摘要
以油菜籽粕为试验材料,以甲醇、乙醇、乙醚、丙酮为提取液,筛选最佳提取溶剂,在单因素的基础上,选定乙醇质量分数、料液比、提取温度和提取时间为自变量,以硫苷得率为响应值,利用BoxBenhnken中心组合设计原理和响应面分析法,研究各自变量的交互作用对硫苷得率的影响,模拟得到二次多项式回归方程的模型。结果表明:在四种提取液中,乙醇提取的硫苷得率较高,且容易浓缩,选择乙醇为最适提取溶剂。通过响应面试验,乙醇溶液质量分数为78%,料液比为1∶8,提取温度63.5℃,提取时间60min,硫苷得率为18.72 mg/g。此研究为从油菜籽粕中提取硫苷提供理论依据。
Rapeseed meal was used as a test material,methanol,ethanol,ethyl ether,and acetone were used as a extract,screening the best extraction solvent,on the basis of the single factor experiment,selected ethanol mass fraction,solid-liquid ratio,extraction temperature and the extraction time were the main independent variables,glycosides yield response using Box-Benhnken central composite design and response surface analysis principles study the effect of each variable interaction of glucosinolates yield,simulated model quadratic polynomial regression equation. Results show that the four kinds of extract,ethanol extract of glucosinolates yield was higher,and easier to concentrate,choose ethanol as the optimum extracting solvent. Through the response surface test, the optimum condition for the extraction of glucosinolates was :ethanol concentration,solid-liquid ratio,extraction temperature and extraction time were 78 %,1 ∶ 8 g/m L,63.5 ℃,and 60 min,respectively. The optimal yield of glucosinolates was18.72 mg/g. The research will provide theoretical support for the anthocyanin of practical application.
