Expression of carbohydrate response element binding protein in hepatocellular carcinoma and its functions

摘要 目的:探讨碳水化合物反应元件结合蛋白(Ch REBP)在肝癌(HCC)中的表达与生物学作用。方法:分别用q RT-PCR、免疫组化、Western blot法检测73例HCC组织与癌旁组织以及多种HCC细胞系与正常肝细胞系中Ch REBP的m RNA与蛋白表达;观察si RNA干扰Ch REBP表达后,HCC细胞周期、凋亡以及增殖的变化。结果:Ch REBP的m RNA与蛋白表达在HCC组织中表达均明显高于癌旁组织、在所有HCC细胞系中均明显高于正常肝细胞系(均P<0.05)。干扰Ch REBP表达后,HCC细胞发生明显G1-S期阻滞、细胞增殖明显降低(均P<0.05),但细胞凋亡未发生明显变化(P>0.05)。结论:Ch REBP在HCC中表达升高,且可能通过调控细胞周期促进HCC细胞的增殖,从而在HCC的发展中起了重要的作用。 Objective: To investigate the expression of carbohydrate response element binding protein (ChREBP) and its biological actions in hepatocellular carcinoma (HCC). Methods: hTe mRNA and protein expressions of ChREBP in 73 specimens of HCC along with its adjacent tissue, as well as different HCC cell lines and normal hepatic cell line were determined by q-PCR, immunohistochemical staining and Western blot analysis, respectively. hTe changes in cell cycle, apoptosis and proliferation in HCC cells were observed atfer the siRNA interference of ChREBP2 expression. Results: Both mRNA and protein expressions of ChREBP were signiifcantly increased in HCC tissue compared with tumor-adjacent tissue, and signiifcantly increased in all studied HCC cell lines compared with normal hepatic cell line (allP〈0.05). After interference of ChREBP2 expression, HCC cells presented significant G1/S phase arrest and inhibition of proliferation (bothP〈0.05), but apoptosis showed no signiifcant change (P〉0.05). Conclusion: ChREBP expression is elevated in HCC, which may probably promote proliferation of HCC cells through cell cycle regulation, and thereby plays an important role in the progression of HCC.
