摘要
对组织滴虫病临床发病鸡的肝和盲肠进行了虫体的分离及体外培养,通过形态学和基于18S rRNA的分子生物学方法进行了鉴定,并采用连续稀释法进行了克隆和建株。结果表明,采用含有马血清和米粉以及抗生素的M 199培养基,从病鸡盲肠中能分离到芽囊原虫、鸡四毛滴虫和火鸡组织滴虫,盲肠中混合感染且难以分离纯化单一虫种;从病鸡肝中能成功分离到火鸡组织滴虫单一虫种;通过有限稀释法成功获得了火鸡组织滴虫的单个克隆虫株,命名为JSYZ-A株。将分离的虫株通过泄殖腔回归接种14日龄黄羽肉鸡,成功复制出了组织滴虫病。本研究结果为进一步开展虫体致病机制、药物筛选和疫苗研制奠定了很好的基础。
A monoclonal strain of Histomonas meleagridis was isolated from clinical sick chickens by cultivation and limited dilution, which was further conformed by morphology and 18S rRNA sequence analysis. Results showed that H. meleagridis could be cultured with M199 culture medium containing horse serum, rice powder and antibiotics. By in vitro cultivation, mixed infections with Blastocystis spp., Tetratrichomonas gallinarum and H. meleagridis were observed in the cecum, while single infection with H. meleagridis was observed in the liver. A monoclonal strain of H. meleagridis named JSYZ-A was obtained with limited dilution. Yellow meat-type chickens were infected intracloacally with JSYZ-A at a dose of 10 000 trophozoites, and histomoniasis was successfully replicated. Results above will lay the foundation for the further studies of pathogenic mechanism, drug screening and vaccine development against H. meleagridis.
