摘要 目的紫外分光光度法测定新疆地产中药红花中总黄酮的含量。方法以芦丁为对照品,以紫外分光光度法(UV)为分析检测方法测定总黄酮的含量。结果用分光光度计进行全波长扫描,在510nm波长处测的最大吸光度,芦丁浓度与吸光度的线性方程为y=12.554x-0.003(R^2=0.9982),表明在8~48μg·m L^(-1)浓度范围内具有良好的线性关系,精密度RSD 0.19%,稳定性RSD 0.14%,重现性RSD 0.11%,平均加样回收率103.3%,RSD为1.08%(n=9)。结论该方法具有较好的精密度,简单易行,科学合理,适用于评价红花药材的质量。 OBJECTIVE To Determinate the content of total flavonoids in Xinjiang safflower of traditional medicine by ultraviolet( UV) Spectrophotometry. METHODS By UV spectrophotometry,the total flavonoids content was determinated,and rutin was used as the control. RESULTS Full wavelength scanning using a spectrophotometer,a wavelength of maximum absorbance was measured at 510 nm,the linear equation of rutin concentration and absorbance was y = 12. 554x- 0. 003( R^2= 0. 9982),indicating a good linear relationship in the concentration range of 8~ 48μg·m L^-1,RSD was 0. 19%,the stability of RSD was 0. 14%,reproducibility RSD was 0. 11%,the average recovery was 103. 3%( RSD = 1. 08%,n = 9). CONCLUSION The method has a good precision,simple,scientific and reasonable,applicable to the evaluation of the quality of safflower.
