核糖核酸腺苷脱氨酶1在肝癌中的表达及临床意义

Expression and influence of adenosine deaminase acting on RNA 1 in primary liver cancer

摘要 目的探讨核糖核酸腺苷脱氨酶1(ADAR1)在原发性肝癌中的表达及其与肝癌临床病理特征的关系。方法免疫组织化学及Western blot法检测ADARl在癌旁组织及肝癌肿瘤组织中表达;收集患者年龄、性别、肿瘤直径、门脉癌栓、肝背膜受侵、分化程度、血清甲胎蛋白(AFP)及细胞核增殖抗原(Ki-67)的表达,了解ADAR1与肝癌临床病理特征的关系。结果免疫组织化学检测结果显示,ADAR1在癌旁组织中ADAR1表达率为29.32%,而在肝癌肿瘤组织中表达率为64.50%,肿瘤组织ADAR1阳性率远高于癌旁组织,差异有统计学意义(P〈0.05);Western blot结果表明,癌组织ADAR1表达量明显高于癌旁组织,差异有统计学意义(P〈0.05);肝癌组织中的ADAR1表达水平与患者年龄、性别、肿瘤直径、门脉癌栓及肝背膜受侵差异无统计学意义(P〉0.05),而与分化程度、血清AFP及Ki-67的表达密切相关,ADAR1表达水平明显升高(P〈0.05)。结论ADARl在肝癌中表达增加,且与肝癌分化程度和血管生成基因相关,提示ADARl可能通过诱导肝癌分化及血管生成而促进肝癌发生侵袭转移。 Objective To investigate adenosine deaminase acting on RNA 1 ( ADAR1 ) in primary liver cancer (PLC) and its relationship with clinical pathology. Methods ADAR1 expression was detected by immunohistochemistry and Western blotting assay in cancer tissues and tumor adjacent tissue; age, gender, tumor size, portal vein tumor thrombus, liver back film invasion, differentiation, serum alpha fetal protein (AFP) and proliferation cell nuclear antigen (Ki -67 ) expression were collected to explore relationship of ADAR1 and clinical pathology. Results Immunohistochemical detection showed that ADAR1 expression rate of tumor adjacent tissue was 29. 3%, and tumor tissue was 64. 50%, there was significant difference of ADARI positive rate between tumor tissue and adjacent tissue ( P 〈 0. 05 ) ; Western blotting assay results showed that ADAR1 expression in cancer tissue was significantly higher than that in the tumor adjacent tissue, the difference was statistically significant ( P 〈 0. 05 ) ; There were no statistical significance between ADAR1 expression levels and patient age, gender, tumor size, portal vein tumor thrombus and liver back film invasion showed ( P 〉 0.05 ) , and the degree of differentiation, serum AFP and Ki - 67 expression is closely related to that ADAR1 expression level increased significantly ( P 〈 0.05 ). Conclusion ADARI expression in PLC increased associated with tumor differentiation and angiogenesis gene, suggesting that ADAR1 may induce degree of differentiation and angiogenesis to promote liver cancer invasion.
