微顺序注射-钙黄绿素荧光猝灭法测定利福平

Establishment the method of determining rifampicin by using micro order flow injection-calce in fluorescence quenching method

摘要 在碱性条件下,利福平对钙黄绿素产生的化学发光信号具有显著的抑制作用,据此结合微顺序流动注射分析技术,建立了快速测定利福平的微顺序注射-钙黄绿素荧光猝灭方法。在碱性介质中,钙黄绿素浓度为33.33μg/m L时其发光强度达到最大值;在此实验条件下,利福平在20.64~103.2μg/m L范围内与钙黄绿素荧光猝灭强度呈良好的线性关系,方程为IF=-192.57ρ+44624(r=0.9997)。 Based on the strong inhibitive effect of rifampicin to chemiluminescence signal of calcein in alkaline medium,micro order flow injection analysis technology was used to establish a method of micro order flow injection with calcein fluorescence quenching for rapid determination of rifampicin. The luminous intensity of calcein was maximum when concentration of calcein was 33. 33 μg / m L in alkaline medium. Under this condition,the concentration of rifampicin in the range of 20. 64 to 103. 2 μg / m L and intensity of calcein fluorescence quenching showed a good linear relationship. The linear equation was Y = 192. 57ρ + 44624( r = 0. 9997). The method of micro order flow injection combined with calcein fluorescence quenching was rapidly,accurately,and well reproducibility with wide linear range. It provided a new basis for determination of content of rifampicin.
