一株日本鳗鲡肠道乳酸菌SOD酶基因克隆表达与应用研究

Cloning and Application of SOD Protein from Lactococcus lactis on the Intestinal of A. japonica

摘要 为了解决日本鳗鲡抗生素滥用的问题,探究能够替代抗生素的微生态制剂,从日本鳗鲡肠道中筛选益生菌乳酸菌,克隆其SOD酶基因,并在大肠杆菌中进行表达,将SOD酶、益生乳酸菌同基础饲料配制成3组不同的饲料饲喂鳗鲡后进行血液生化指标分析.实验结果表明,益生菌、SOD酶以及乳酸菌和SOD酶制成的复合酶饲养的鳗鲡的血清总蛋白相对于对照组分别提高2%、8%、18%;葡萄糖相对于对照组分别提高64.8%、19.1%、79.7%;超氧化物歧化酶相对于对照组分别提高10.7%、25.4%、47.1%.各项指标提示添加乳酸菌、SOD酶蛋白有利于日本鳗鲡的生长,在一定程度上提高宿主的抗病能力和免疫性能. In order to solve the problem of antibiotics abuse in aquaculture of Anguilla japonica and search for the probiotics which can replace antibiotics,a probiotic strain named Lactococcus lactis was screened from the intestinal of A. japonica,and a SOD gene was cloned from L. lactis and expressed in Escherichia coli. The blood biochemical parameters of A. japonica in three groups fed respectively with SOD protein,L. lactis cells,and SOD plus the probiotic cells,which were all mixed with basic food respectively,were analyzed. The results showed that the measure of the total serum protein in the probiotic groups,SOD group and the group of SOD combined with probiotic cells were increased by 2 %,8 %,and 18 % respectively in comparison with the control group.The measure of glucose increased by 64. 8 %,19. 1 %,and 79. 7 % respectively. The activity of superoxide dismutase increased by 10. 7 %,25. 4 %,and 47. 1 % respectively. All of the indicators suggested that the addition of L. lactis cells and SOD protein in the food were beneficial to the growth of A. japonica and could improve its immunity and resistance to diseases.
