金钗石斛总生物碱的提取条件优化 被引量:7

摘要 为科学合理开发金钗石斛药材资源,采用单因素试验分析提取温度、乙醇浓度、提取时间、固液比和提取次数对金钗石斛总生物碱含量的影响,酸碱滴定法测定总生物碱含量。在此基础上,运用正交试验对乙醇浓度、提取时间、固液比和提取次数进行优化。结果表明:金钗石斛总生物碱提取制备的最佳条件为提取温度85℃,乙醇浓度90%,提取时间3h,固液比1∶20,提取次数为2次,在该条件下金钗石斛总生物碱的平均提取率为87.8mg/g。 In order to scientifically and rationally exploit medicinal resources of D.nobile,a singlefactor experiment was conducted to analyze the effects of extraction temperature,ethanol concentration,extraction time,solid-liquid ratio and extraction times on total alkaloids from D.nobile,and the acid-base titration method was applied in determination of total alkaloids.On the basis of this test,an orthogonal experimental design test was applied to further investigate the extraction process for total alkaloids from D.nobile,the concentration of ethanol aqueous solution,extraction time,solid-liquid ratio and times of extraction were reviewed seriously.Results:The optimum extraction process of total alkaloids from D.nobileincluded extraction temperature 85℃,90% ethanol concentration,extraction time 3h,solid-liquid ratio 1∶20g/mL,two times of extraction.The average extraction rate under this optimum condition for total alkaloids from D.nobile was 87.8mg/g.
