增龄对内皮祖细胞数量和功能活性的影响及与血管内皮功能的关系 被引量:6

摘要 目的探讨增龄对人外周血内皮祖细胞数量和功能活性的影响及与血管内皮功能的关系。方法入选32例健康老年人作为实验组(老年组)和30例健康青年人为对照组(青年组)。抽取受试者外周静脉血20 ml,采用密度梯度离心法分离提取内皮祖细胞,采用流式细胞技术比较两组志愿者循环中内皮祖细胞数量。体外培养7 d后,分别检测两组志愿者内皮祖细胞体外迁移及黏附能力;通过裸鼠颈动脉拉脱模型,比较内皮祖细胞在体内皮损伤修复能力。检测两组志愿者血流介导的血管舒张功能(FMD)。结果与青年组相比,老年组外周血中内皮祖细胞数量明显下降(P<0.01),体外迁移、黏附(P<0.01)及在体再内皮化功能均明显受损。减少的在体再内皮化面积与下降的FMD之间呈正相关。结论增龄导致外周血中内皮祖细胞数量和功能活性受损与血管内皮功能下降密切相关,是动脉粥样硬化血管疾病发生发展的重要环节。 Objective To investigate the effect of aging on the quantity and functional activities of circulating endothelial progenitor cells( EPCs) in healthy men. Methods 32 healthy old people〔elderly group:( 68 ± 6. 6) years old〕and 32 healthy young people〔young group:( 26 ± 5. 8) years old〕were enrolled into the research. EPCs were isolated and cultured from peripheral blood by density gradient centrifugation method,and the quantity of circulating EPCs was exterminated by flow cytometry analysis. Further,the migration and adhesion function in vitro and the reendothelialization capacity in vivo of EPCs from two groups subjects were measured. Moreover,the flow-mediated dilatation of two group subjects were exterminated. Results Compare with those of young group,the quantity( P〈0. 01),in vitro migration and adhesion( P〈0. 01) and in vivo reendothelialization capacity( P〈0. 001) of EPCs from aging group were significant declined. And the impaired endothelial repair capacity was positive relation with declined FMD. Conclusions Age-related decline in quantity and function of human EPCs is close relation with decreasing endothelial function,which is an important risk factor for atherosclerotic vascular diseases.
