复方苦黛汤不同提取部位抗炎作用的筛选及作用机制初探 被引量:3

摘要 目的探讨复方苦黛汤不同提取部位对溃疡性结肠炎(UC)大鼠的治疗作用及其作用机制。方法通过观测UC大鼠一般情况和病理组织切片HE染色,检测复方苦黛汤不同提取部位对TNBS诱导的UC大鼠结肠组织髓过氧化物酶(MPO)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)和一氧化氮合酶(NOS)的影响,评价其不同提取部位对UC大鼠的治疗作用。结果复方苦黛汤氯仿提取部位可明显降低UC大鼠结肠组织中MPO、MDA、NOS和NO水平,增加SOD和GSH-Px活性,同时大鼠一般情况及病理组织HE染色亦显著得到改善。结论复方苦黛汤氯仿提取部位能显著抑制UC大鼠的氧化应激反应,减少结肠炎症损害,可以筛选复方苦黛汤的氯仿部位作为进一步的研究分析。 Objective To investigate the curative effect and mechanism of action of different extracts from Fufang Kudai decoction on ulcerative colitis(UC) in rats. Methods We observed the general condition and pathological biopsy in rats with UC HE staining, examined the effect of different extracts from Fufang Kudai decoction on TNBS induced UC rats colon tissues, including myeloperoxidase(MPO), malondialdehyde(MDA), superoxide dismutase(SOD), glutathiane peroxidase(GSH-Px), nitric oxide(NO) and nitric oxide synthase(NOS), and evaluated the curative effect in UC treatment in rats. Results Fufang Kudai decoction chloroform extract obviously reduced the MPO and MDA, NOS, and NO level in UC rats colon tissues, increased SOD and GSH-Px activity, and at the same time significantly improved the general situation of rats and histopathologic HE staining. Conclusion Fufang Kudai decoction chloroform extract can significantly inhibit the oxidation of UC stress reaction in rats, and reduce the colon inflammation damage. Fufang Kudai decoction chloroform parts is worth further research.
