摘要
目的:探讨验方煎剂中有效成分的影响因素。方法:利用高效液相色谱技术,以选定的验方药材中芍药苷和党参炔苷含量为评价指标,分析提取温度、提取溶剂、提取方法、药材配伍等因素对其的影响。结果:50%乙醇、60℃水浴加热结合微波辅助法对有效成分的保留效果最好,芍药苷、党参炔苷在验方中的含量分别为42.15、0.67 mg/g,在单一药材赤芍和党参中的含量分别为39.99、0.68 mg/g。60℃以上水浴温度不利于芍药苷成分保留。药材配伍对党参炔苷含量的影响较小,但对芍药苷含量的影响显著。结论:在验方煎制过程中,采用有机溶剂提取、适当水浴加热及微波辅助法,有利于保留其有效成分。不同的提取对象所受到的影响因素及效果各不相同,煎制时应综合考虑药材配伍、提取对象理化性质、酸碱度等多种因素。
OBJECTIVE:To probe into the effect factors of active ingredient extraction in empirical prescription . METHODS:High-performance liquid chromatography ( HPLC ) were adopted , to set the content of paeoniflorin and lobetyolin in the selected prescription medicine as evaluation indicators , effects of extraction method , temperature , solvent and compatibility on active ingredient extraction were studied .RESULTS:50%ethanol solution and 60℃water path combined with microwave method could extract the highest rate of active ingredient .Contents of paeoniflorin and lobetyolin in the selected prescription medicine were respectively 42.15 mg/g and 0.67 mg/g;which were 39.99 mg/g and 0.68 mg/g in radix paeoniae rubra and codonopsis respectively .Temperature over 60 ℃was not conducive to the extraction of active ingredient .Compatibility had greater influence on paeoniflorin extraction than that on lobetyolin . CONCLUSIONS:During the process , organic solvent extraction , appropriate water bath heating and microwave method are helpful for the increase of extraction efficiency of active ingredient .Different extraction objections can be influenced by different factors and the effects are different , therefore , it is necessary to comprehensively take the compatibility , physic-chemical property of objects and pH into consideration .
