摘要
应用冰冻切片法观察艾纳香成熟叶的显微结构。结果显示,优化的冰冻切片方法为:取新鲜叶片,不经预处理,直接切取4 mm×4 mm小块,用冷凝胶包埋,冷凝箱温度和样品夹温度-18°C,冷凝时间10 min,修片后取出并置于室温20℃条件下回温30 s,移回样品夹后30 s内切片,切片厚度20μm,用洁净载玻片吸片,于光镜下观察。成熟艾纳香叶为异面叶,厚度约110μm;栅栏组织和海绵组织分化明显,栅栏组织由1层排列紧密的柱状细胞组成,海绵组织有发达的细胞间隙,两者之比为0.82~1.22;叶中脉发达,主脉散生,外韧维管束6~10个,由厚壁细胞组成维管束鞘,导管为螺纹;叶上、下表皮均有大量非腺毛和腺毛分布,叶表皮的腺毛为双列多细胞组成的头状腺毛。本研究结果确定了利用冰冻切片研究艾纳香叶显微结构简便可行,为艾纳香属植物分类鉴定、品种整理等提供参考,并为进一步研究艾纳香的结构和发育积累经验。
This paper studies the microstructure of sambong's leaf by freeze-sectioning. The optimized method is to collect a fresh and healthy adult leaf and cut a piece from it with an area of 4 mmx4 mm without pretreatment. Then the piece is embedded with Jung tissue freezing medium under -18 ℃ for 10 min, and then placed under room temperature at 20 ℃ for 30 seconds and cut under -18℃ within 30 s. The section thickness is 20 μm. A glass slide is used to stick the section. The section is observed with a optical microscope. The results showed that the ainaxiang leaf is typically bifacial and is with a thickness of about l 10 μm. Palisade tissue and spongy tissue in leaves are differentiated. Palisade tissue is composes of columnar cells closely arranged one layer. Intercellular space of spongy tissue is developed. The ratio of palisade tissue thickness to spongy tissue thickness ranges from 0.82:1 to 1.22: 1. Midrib is developed. Six to eight collateral vascular bundles are loosely arranged in midrib. Vascular bundle is consists of spiral vessels inside and sclerenchyma cells around as vascular bundle sheath. Both non-glandular hairs and glandular hairs are occurred frequently in both sides of adult leaves. The glandular hairs are capitate trichomes consists of several cells arrange in two lines. Above results confirm that the feasibility and convenience of studying ainaxiang leaf microstructure with freezing sections. This finding may help better taxonomical study of Blumea plants, and add some information for further study on microstructure and development of this plant.
