摘要
目的建立高效液相色谱法同时测定大鼠血浆中紫杉醇棕榈酸酯(PTX-PA)及其活性代谢产物紫杉醇(PTX)含量。方法以多西他赛作为内标,以乙腈和水作为流动相进行梯度洗脱,流速为1m L·min-1,检测波长227nm,柱温30℃。结果紫杉醇棕榈酸酯(r=0.9993)、紫杉醇(r=0.9999)在0.1-50μg·m L-1范围内线性关系良好。紫杉醇棕榈酸酯、紫杉醇定量限分别为100ng·m L-1、50ng·m L-1,两组分提取回收率均大于80%,两组分日内、日间精密度RSD%均小于5%。结论本方法快速、简便、准确可靠,适合于大鼠血浆中紫杉醇棕榈酸酯及其活性代谢产物紫杉醇的检测。
OBJECTIVE To establish the method for the simultaneous determination content of paclitaxel palmitate( PTX-PA) and its active metabolite paclitaxel( PTX) in rat plasma by HPLC. METHODS With docetaxel as the internal standard,the chromatographic separation was performed on an ODS-2 C18column( 250 × 4. 6mm,5. 0μm particle size) with gsadient elution by using acetonitrile-water as the mobile phase with UV detection at 227 nm,the flow rate was 1m L·min- 1and the column temperature was 30℃. RESULTS The method showed a good linearity in the range of 0. 1 - 50μg·m L- 1for paclitaxel palmitate( r = 0. 9993) and paclitaxel( r = 0. 9999),while the limits of quantitation were 100 ng · m L- 1and 50 ng · m L- 1,respectively. The extraction recoveries of the two ingredients were more than 80%. Moreover,the RSD for inter-day assay and intra-day assay were less than 5%. CONCLUSION The method is rapid,simple,accurate and reliable,therefore it is suitable for the detection of paclitaxel palmitate and its active metabolite paclitaxel in rat plasma.
