摘要
基因测序技术已发展至第四代,其中第二代、第三代和第四代测序技术统称为下一代测序(NGS)技术。基因测序原理也相应地经历了Sanger测序法,边合成边测序、单分子测序和纳米孔测序。NGS技术因为通量提高、成本降低和测序周期缩短的优势已被广泛应用于基因组学、转录组学、宏基因组学和表观组学等方面。本文就NGS技术的基本原理和应用特点进行综述。
Gene sequencing technology has been developed into the fourth generation, the second, the third and the fourth generations are collectively referred to as next generation sequencing(NGS) technologies. The corresponding principle of gene sequencing has experienced the Sanger sequencing, sequencing by synthesis,single molecule sequencing and nanopore sequencing accordingly. NGS technologies have been widely applied to genomics, transcriptomics, metagenomics and epigenetics for the increased throughput, reduced cost and shortened sequencing cycle. In this article, the basic principles and application features of NGS technologies were reviewed.
