地榆皂苷类成分对环磷酰胺致小鼠骨髓抑制的保护作用研究 被引量:14

摘要 为了观察地榆皂苷类成分对骨髓抑制的保护作用,本文采用腹腔注射环磷酰胺致小鼠骨髓抑制模型。造模成功后,将骨髓抑制小鼠按体重随机分为:模型对照组、集落刺激因子组、地榆皂苷Ⅰ5、2.5、0.5 mg/kg三个剂量组,地榆皂苷Ⅱ5、2.5、0.5 mg/kg三个剂量组,地榆皂苷元5、2.5、0.5 mg/kg三个剂量组,另设正常对照组。连续灌胃7 d,每天一次,以外周血液白细胞数量、骨髓细胞DNA含量和CD34+的表达以及骨髓细胞O6-甲基鸟嘌呤-DNA甲基转移酶(MGMT)蛋白的表达为评价指标,研究地榆皂苷类成分对CTX所致骨髓抑制的治疗作用。结果显示地榆皂苷类成分可显著升高小鼠WBC(P<0.05);显著升高小鼠骨髓DNA含量(P<0.05);显著促进CD34+的表达(P<0.05);显著促进MGMT蛋白的表达(P<0.05)。通过本研究可以看出地榆皂苷类成分可显著拮抗CTX所致小鼠骨髓抑制。 The objective of this study was to investigate the protective effect of tannins from Sanguisorba officinalis on myelosuppression in mice. The myelosuppression model was induced by peritoneal injection of cyclophosphamide (CTX). Then the myelosuppression mice were randomly divided into: model group, colony-stimulating factor group, three ziyuglycoside-I treatment groups (5,2.5,0.5 mg/kg), three ziyuglycoside- 11 treatment groups (5,2.5,0.5 mg/kg), three Bumet prosapogenins treatment groups (5,2.5,0.5 mg/kg) and normal group. The drugs were chronically admin- istrated to myelosuppression mice once a day for 7 days. The counts of peripheral blood leukocytes, D1NA contents and CD34+ expression of bone marrow cells and the O6-methyl guanine-DNA methyltransferase (MGMT) protein expression were used as main indexes and were determined. The results showed that S. officinalis significantly increased peripheral WBC ( P 〈 0.05) ; significantly elevated bone marrow DNA content (P 〈 0.05) ; significantly promoted the expression of CD34 + ( P 〈 0.05 ) ; also significantly promoted MGMT protein expression ( P 〈 0.05 ). Based on these results, it was concluded that S. offwinalis can significantly antagonize CTX-induced myelosuppression in mice.
