摘要
目的:观察白蛋白结合型紫杉醇(nab-PC)对宫颈癌Caski细胞凋亡的影响并探讨其可能的机制。方法:将30只实验用615小鼠随机分为对照组和nab-PC组,于右腋窝下皮下接种0.1m L宫颈癌Caski细胞,连续15d。对照组右腋窝皮下注射0.9%氯化钠注射液0.2m L,nab-PC组右腋窝皮下注射nab-PC 0.2m L,均每周2次,治疗3周。采用免疫组化技术检测两组肿瘤组织中Bcl-2、Bax及Caspase-9的蛋白表达水平。结果:与对照组相比,nab-PC组Bcl-2蛋白表达水平明显降低,Bax、Caspase-9蛋白表达水平及Bax/Bcl-2比值明显升高,差异均有统计学意义(P<0.05)。结论:白蛋白结合型紫杉醇具有较强的促进宫颈癌Caski细胞凋亡的作用,其机制可能与增加Bax/Bcl-2比值和激活Caspase-9等有关。
Objective: To investigate the effect of nanoparticle albumin- bound paclitaxel( nab- PC) on the apoptosis of cervical cancer Caski cells and possible mechanism. Methods: A total of 30 experimental 615 mice were randomly divided into control group and nab- PC group,and subcutaneous inoculation of 0. 1 ml cervical cancer Caski cells in the right axilla were performed for 15 days continuously. The mice in the control group were given subcutaneous injection of 0. 9% sodium chloride 0. 2 ml in the right axilla,and those in the nab- PC group were given subcutaneous injection of nab- PC 0. 2 ml in the right axilla; the treatment was performed twice a week for 3 weeks in both groups. Immunohistochemistry was used to measure the protein expression of Bcl- 2,Bax,and caspase- 9 in tumor tissues in the two groups. Results: Compared with the control group,the nab- PC group showed a significant reduction in the protein expression of Bcl- 2 and significant increases in the protein expression of Bax and caspase- 9 and Bax /Bcl- 2 ratio( P〈0. 05). Conclusion: The nab- PC has a strong effect in promoting the apoptosis of cervical cancer Caski cells,and its mechanism of action may be related to an increased Bax / Bcl- 2 ratio and activation of caspase- 9.
