免疫性血小板减少症的免疫球蛋白序列分析

Analysis of immunoglobulin genes derived from immune thrombocytopenia

摘要 目的:通过分析免疫性血小板减少症(ITP)患者幼稚B细胞免疫球蛋白序列的特点,探索ITP患者B细胞中枢免疫耐受机制的情况。方法:采用单细胞PCR的方法从6例ITP患者和4例健康对照者中克隆并测序了400多个免疫球蛋白基因序列,并借助IMGT数据库、IgBLAST数据库以及VHRFA-1软件等进行分析。结果:与正常者相比,ITP患者的免疫球蛋白轻链序列倾向于运用下游的Vκ和上游的Jκ,免疫球蛋白重链包含更多的VH替换产物,并且其互补决定区(CDR3)中含有更多的正电荷。结论:ITP患者B细胞中枢免疫耐受机制出现紊乱,这可能是ITP发生的机制之一。 Objective:To explore the status of central B cell tolerance in immune thrombocytopenia (ITP) by analyzing the immunoglobulin fig) repertoire in naive B cells. Method:We cloned and sequenced more than 400 Ig genes derived from 6 ITP patients and 4 healthy controls. The sequences were analyzed by programs including IMGT,IgBLAST,and VHRFA-1. Result:The Ig light chains preferentially used downstream Vκ and upstream Jκ. The frequencies of VH replacement products were increased in ITP. Furthermore, the IgH CDR3 regions were en- riched with positively charged amino acids. Conclusion:These features indicate defective central B cell tolerance in ITP,which might be one of the underlying mechanisms of ITP pathogenesis.
