MicroRNA-499b通过亚甲基四氢叶酸还原酶调节老年慢性心力衰竭患者血清同型半胱氨酸水平的机制及意义 被引量:6

摘要 目的探究MicroRNA(MiR)-499b在老年慢性心力衰竭(CHF)患者中的表达情况及其对血清同型半胱氨酸(Hcy)含量的影响。方法利用老年CHF患者的组织芯片,通过原位杂交检测MiR-499b mRNA表达水平;利用荧光素酶和Western印迹检测MiR-499b对其靶基因亚甲基四氢叶酸还原酶(MTHFR)的表达调控;通过检测血清中的Hcy表达情况,显示MiR-499b和MTHFR对Hcy逆转产生的影响;利用荧光素酶和Western印迹实验检测MiR-499b对肿瘤坏死因子(TNF)-α的表达调控,显示MiR-499b和TNF-α对CHF逆转产生的影响。结果 MiR-499b在老年CHF患者中显著降低,并且正向调控MTHFR的表达水平;MiR-499b直接靶向调控MTHFR启动子活性,增高MTHFR的蛋白表达水平,下调Hcy的表达水平,并且发现MiR-499b能够靶向调控TNF-α,降低其表达水平,进而CHF炎症水平,降低心衰发生率。结论 MiR-499b的低表达与血清Hcy的异常表达具有显著相关性,MiR-499b通过增强MTHFR的表达降低血清Hcy的表达量;MiR-499b能够降低TNF-α的表达水平,减少CHF的发生。
