闽西地区猪圆环病毒2型血清学调查及遗传变异分析 被引量:16

Serological investigation and genetic diversity of porcine circovirus type 2 infection in swine farms of western Fujian province

摘要 为了解闽西地区2013-2015年猪圆环病毒2型(PCV2)血清学及流行毒株的遗传变异规律,本研究采集闽西地区规模化猪场1 771份血清样品,应用ELISA方法对其进行PCV2抗体水平检测,结果血清样品的总阳性率为93.00%,其中种猪血清阳性率最高为99.78%,哺乳仔猪、保育猪和育肥前期猪血清阳性率逐渐降低,分别为91.62%,80.78%,77.68%,育肥后期猪血清阳性率上升为100.00%。应用PCR方法对10株PCV2全基因组进行遗传变异分析,结果表明,PCV2基因组全长为1 766-1 768bp,10株PCV2的核苷酸序列同源性为94.6%-99.7%,其与国内外参考毒株的同源性为94.0%-99.2%。系统进化树结果表明,10株PCV2毒株可分为2个大支,1株属于PCV2a,9株属于PCV2b(2株为1A/AB,7株为1C);氨基酸序列分析表明在Cap蛋白的53-91,121-151和185-215位存在3个高度变异区,表明闽西地区PCV2流行毒株主要为PCV2b亚型。 To understand popularity and genetic diversity of porcine circovirus type 2(PCV2)infection in swine farms of the western Fujian province.During 2013 to July 2015,1 771 serum specimens were sampled from large-scale pig farms in Longyan city of western Fujian province to detect the antibody against PCV2 by indirect ELISA.The results showed that the mean PCV2 antibody positive rate of samples was 93.00%,and among these samples,antibody positive ratio of breeding pigs(sows and boars),nursery pigs,piglets,fatteners were 99.78%,91.62%,80.78% and77.68%-100.00%,respectively.Sequencing of the 10 complete genomes of PCV2 showed that the genome lengths differed from 1 766-1 768 bp,and they shared homology of 94.0%-99.2% with different reference PCV2 isolates and 94.6%-99.7% among themselves.The phylogenetic tree analysis showed that 10 isolated strains were divided into two group,PCV2a(1isolate)and PCV2b(2of 1A/AB and 7of 1C).Meanwhile,three major heterogenic regions were observed in amino acid positions 53-91,121-151 and 185-215.These results showed that PCV2 bwere the dominant viruses in western Fujian province.
