菊芋酶解高果糖浆单因素及响应面优化工艺研究 被引量:1

摘要 利用菊粉酶对菊粉进行加工可以高效实现菊粉在食品加工各领域的低成本、多应用、高附加值。采用酶解法水解菊粉,通过底物浓度、适宜温度、pH、酶用量单因素的检测,采取响应面优化生产工艺条件。单因素结果为酶粉浓度35%、50℃条件下调浆30min、pH值5.5~6、加入2mL氯化钙溶液(0.1moL/L),60℃恒温水浴中加入2mg菊粉酶(6 000U)水解40min为单因素最优条件。正交优化后工艺参数为底物浓度34.7%、pH值6.0、水解温度55.5℃、酶用量2.6mg。对水解效果影响的大小顺序是底物浓度〉酶用量〉水解温度〉pH值。 Biotechnological processing of inulin with inulinase can provide an efficient approach for low-cost,multi-directions,and high-valueadded development and utilization of inulin in the field of food.Enzymatic hydrolysis method is adopted in this paper by the single factor of subtrate concentration、temperature、pH value、dosage of inulinase,and the response surface analysis to optimize the process conditions.The best single factor test is that the substrate concentration is 35%,the temperature is 50℃、pH value is 5.5-6,and add 2mL of calcium chloride solution(0.1mol/L)and the 60℃with the dosage of inulinase is 2mg(6 000U).The Orthogonal optimized process parameters results showed as follows:the substrate concentration is 34.7%,pH is 6and the temperature is 55℃ with the dosage of inulinase is 2.6mg.Effect of the hydrolysis performance is substrate concentration〉 dosage of inulinase〉temperature 〉pH value.
