摘要
目的:观察Runx2和Osx在牙髓损伤后表达模式的变化。方法:取雄性SD大鼠24只随机抽取4只不作任何处理,用于正常对照;其余20只分别以双侧下颌第一磨牙为对象建立牙髓损伤模型。分别于建模后0、3、12 h和3、7 d各时间点从牙髓损伤组中各随机抽取4只大鼠,并取其双侧下颌第一磨牙标本制作组织切片;然后采用HE染色法观察各组修复性牙本质的形成情况,并采用免疫荧光染色法检测Runx2、Osx、DSPP的表达模式。结果:HE染色显示,损伤后7 d,在损伤下方可见明显的修复性牙本质形成。免疫荧光染色显示,Runx2、Osx和DSPP主要表达于损伤下方,其中Runx2的表达呈"抛物线"趋势,即损伤后3 d时表达最为强烈(P<0.05),此后逐渐下降;Osx和DSPP的表达均呈逐渐上调的趋势,Osx的表达在损伤后7 d时上调最为明显(P<0.05),而DSPP的表达则在损伤后3 d时即显著增强并持续至损伤后7 d(P<0.05)。结论:Runx2、Osx和DSPP在牙髓损伤后的不同时期具有不同的表达模式。
AIM: To observe the expression of Runx2,Osx and DSPP after dental pulp injury in SD rats.METHODS: 24 male SD rats were randomly divided into control( n = 4) and dental pulp injury groups( n = 20). The pulp of the bilateral mandibular first molars in injury model groups was exposed. At 0 h,3 h,12 h,3 d and 7 d after dental pulp exposure the animals( n = 4) were perfused to death. The teeth were sectioned and the expression of Runx2,Osx and DSPP was observed using HE staining and immunofluorescence. RESULTS: Reparative dentine formation was detected at the 7th after pulp injury. The expression of Runx2,Osx and DSPP was mainly at the bottom of dental pulp injury site. The expression of Runx2 was the strongest at the 3rd after injury( P〈0. 05) and then gradually declined. The expression of Osx and DSPP showed a trend of gradual increase. The expression of Osx was the strongest at the 7th after injury.( P〈0. 05),while the expression of DSPP reached peak at the 3rd after injury and remained constant( P〈0. 05). CONCLUSION: Runx2,Osx and DSPP show different expression patterns in different periods after dental pulp injury.
