摘要
目的探讨黄芩苷对单侧输尿管梗阻(UUO)模型大鼠肾脏氧化应激的影响。方法将90只大鼠随机分为假手术组、模型组、贝那普利组、黄芩苷组及黄芩苷+贝那普利组,每组18只。假手术组大鼠仅切开腹腔并游离左侧输尿管,但不结扎和离断,模型组和各干预组大鼠制备UUO模型。造模后贝那普利组大鼠给予贝那普利混悬液灌胃10 mg·kg^(-1)·d^(-1),黄芩苷组大鼠给予黄芩苷溶液灌胃40 mg·kg^(-1)·d^(-1),黄芩苷+贝那普利组大鼠给予黄芩苷溶液灌胃40 mg·kg^(-1)·d^(-1)和贝那普利混悬液10 mg·kg^(-1)·d^(-1),模型组与假手术组大鼠给予等量生理盐水灌胃。饲养14、21、28 d后处死各组大鼠,取肾脏标本,用可见光分光光度计法测定肾组织中超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-PX)的含量;酶联免疫吸附试验测定晚期氧化蛋白产物(AOPP)的含量。结果与假手术组比较,模型组各时间点大鼠肾组织中SOD、GSH-PX活性及T-AOC均降低(P<0.05),AOPP含量升高(P<0.05)。与模型组比较,黄芩苷组、贝那普利组和黄芩苷+贝那普利组各时间点大鼠肾组织中SOD、GSH-PX活性及T-AOC均显著升高(P<0.05),AOPP含量降低(P<0.05)。黄芩苷组、贝那普利组和黄芩苷+贝那普利组3组之间各时间点大鼠肾组织中SOD、GSH-PX活性、T-AOC及AOPP含量两两比较差异均无统计学意义(P>0.05)。结论黄芩苷对UUO模型大鼠肾脏氧化应激具有保护作用。
Objective To observe the protective effect of Baicalin on oxidative stress in rats with unilateral ureteral obstruction( UUO). Methods Ninety Sprague-Dawley male rats were randomly divided into sham operation group,model group,benazepril group,Baicalin group,Baicalin + benazepril group,with 18 rats in each group. The UUO model was established in the model group and the intervention group rats. In sham operation group,the rats underwent abdominal incision and free left ureter,but not ligation and amputation. After modeling,the rats in benazepril group received intragastric administration of benazepril suspension 10 mg · kg^(-1)· d^(-1); the rats in Baicalin group received intragastric administration of Baicalin40 mg·kg^(-1)·d^(-1); the rats in Baicalin + benazepril group received intragastric administration of Baicalin 40 mg·kg^(-1)·d^(-1)and benazepril suspension 10 mg·kg^(-1)·d^(-1); the rats in model group and sham operation were given the same volume of normal saline. Six rats were sacrificed in each group on the 14 th,21th,28 thday after experiment onset and the kidney were obtained. The visible range spectrophotometer method was used to determine the activity of superoxide dismutase( SOD) and glutathion peroxidase( GSH-PX),and the total antioxidant capacity( T-AOC) in kidney tissue. The enzyme linked immunosorbent assay was used to determine the contents of advanced oxidation protein products( AOPP). Results Compared with the sham operation group,the activity of SOD and GSH-PX,and the T-AOC level in model group decreased( P 0. 05) and the contents of AOPP increased( P 0. 05) in each time point. Compared with the model group,the activity of SOD and GSH-PX,and the T-AOC level in Baicalin group,benazepril group and Baicalin + benazepril group increased( P 0. 05) and the contents of AOPP decreased( P 0. 05) in each time point.
