摘要
目的:考察超声波辅助乙醇提取法对东北茶藨子叶片中总酚、总黄酮提取含量的影响。方法:采用比色法测定总酚、总黄酮的含量。在单因素实验的基础上,通过Box-Behnken实验设计(BBD)结合响应面法(RSA)优化提取条件,并对不同提取方法进行了比较。结果:超声波法提取东北茶藨子叶片中总酚(以没食子酸计)、总黄酮(以芦丁计)的最佳提取条件为乙醇浓度70%、液固比24 m L/g、超声时间30 min、超声温度40℃、超声功率200 W,在此条件下东北茶藨子叶片中总酚、总黄酮含量分别为135.80 mg/g和13.56 mg/g。结论:超声辅助乙醇提取法高效、快速,可以作为东北茶藨子叶片中总酚、总黄酮的提取方法。
Objective:Ethanol ultrasonic-assisted extraction of total phenols and flavonoids from Ribes mandshuricum(Maxim.)Kom leaves was investigated to optimumize the extraction conditions.Method:Colorimetric method was used to determine the total content of phenols and flavonoids.Based on the single factor,we used Box-Behnken Design combined with response surface analysis to optimize extraction conditions,finally compared the different extracting methods.Results:The optimum condition of extracting total phenols(Gallic acid/g extract) and flavonoids(rutin/g extract) was as follows:70% ethanol,ratio of liquid to material of 24 m L/g,at40℃,under 200 W,for 30 min.The yields of total phenols and flavonoids could reach 135.80mg/g and 14.56mg/g,respectively.Conclusion:With its high efficiency as well as rapid extracting speed,Ultrasound-assisted alcohol extraction method is suitable to extract total phenols and flavonoids from Ribes mandshuricum(Maxim.)Kom leaves.
