摘要
目的建立高效提取盾叶薯蓣中薯蓣总皂苷的新方法。方法以薯蓣总皂苷的得率为指标,采用单因素试验对生物酶预处理过程的酶解温度、pH、生物酶预处理水用量、酶用量、酶解时间5个因素进行考察,以正交试验设计对醇提总皂苷过程的乙醇浓度、料液比、提取时间、提取次数4个因素进行考察。结果生物酶预处理过程最佳酶解条件:酶解温度70℃,pH=5.5,生物酶预处理水用量4 L/kg,酶用量8 m L/kg,酶解时间24 h;醇提总皂苷过程最佳提取条件:60%乙醇为溶剂,料液比1∶8,提取2次,每次1.0 h。结论新工艺可稳定高效地提取出盾叶薯蓣中的薯蓣总皂苷。
Objective To establish a highly efficient extraction method for the saponins extracted from dioscorea zingiberensis. Methods Evaluated by the yield of the saponins, the 5 factors, namely temperature, pH, water consumption, dosage of enzyme and enzyme hydrolysis time in the process of enzyme pretreatment were optimized via single factor experiment, and the 4 factors, namely ethanol concentration,ratio of solid to liquid, extracted time and extraction degree in the saponins extracion process were optimized via orthogonal experiment. Results The best conditions in the enzyme pretreatment are as follows: the temperature for the enzyme was 70 ℃, pH 5. 5, water consumption was 4 L/kg, dosage of enzyme was 8 mL/kg, enzyme hydrolysis time was 24h; the best conditions in the saponins extracted process were as follows: ethanol concentration 60%, ratio of solid to liquid 1: 8, extraction time 1.0h, extraction degree twice. Conclusion The optimal conditions can extract the saponins from dioscorea zingiberensis stably.
