摘要
目的 采用去卵巢大鼠模型 ,观察服用他汀药 (simvastatin ,舒降之 )与二膦酸盐药物(etidronate ,邦德林 )对股骨下端骨量影响 ,进一步探讨他汀药物的治疗骨质疏松的疗效机制。方法 切除卵巢大鼠在术后 1个月开始分别给予辛伐他汀与二膦酸盐及对照激素替代治疗 ,同时设假手术对照与去势对照组。各组均在服药 10 0d后处死动物 ,进行全股骨骨密度及股骨下端不脱钙制片的骨组织形态计量学观察。结果 服药组均较去势对照组的骨密度值有所增加。骨计量学测量同样可见到骨体积明显改善。他汀类药物主要作用于骨形成类指标 ,如 :成骨活性增加、类骨质生成、骨转换加快、骨重建时间缩短等 ,这均与二膦酸盐药物作用机制与表现不同。二膦酸盐作用使骨吸收表面减少、骨重建周期延长 ,而类骨质表面与平均类骨质宽度等计量指标下降。结论 他汀类药物能阻止去卵巢大鼠股骨下端骨组织的快速骨丢失 ,其作用是通过促进骨组织形成与骨转换加快而实现 。
Objective\ To observe the effects of simvastatin and bisphosphonates on distal femur of ovariectomized rats and study the anti osteoporotic mechanism of statins. Methods\ One month after removal of ovaries from rats, administration of statins, bisphosphonates and hormone substitute therapy began, and simultaneously sham operated and ovariectomized rats were used as controls. One hundred days after the treatments, the rats were killed, bone mineral densities(BMD) were measured, and bone histomorphometry of undecalcified sections was porformed. Results\ BMD in the treated groups increased, compared with the untreated ovariectomized rats; bone volume was improved significantly. The statins mainly influenced the indices of bone formation, such as the formation of osteoid, increasing osteoblast activity and bone turnover and shortening the period of bone rebuilding, etc; all of them were different from the action and mechanism of bisphosphonates. Conclusion\ Through increasing bone formation and bone turnover, statins can prevent the rapid loss of the distal femur of ovariectomized rats, which is different from the action of bisphosphonates.\;
