摘要
目的评估基于VP4区基因序列的肠道病毒分型方法准确性,建立快速简便的肠道病毒分型方法。方法应用逆转录-聚合酶链反应(RT-PCR)对126株已用VP1区基因序列完成分型鉴定的肠道病毒进行VP4区基因扩增,对扩增产物测序分析,获得VP4区分型结果。结果 126株肠道病毒经VP4区基因序列分型共分出30个型别,包括肠道病毒A组37株8个型别,肠道病毒B组85株20个型别,肠道病毒C组2株2个型别。与VP1区分型结果比较,A、C组肠道病毒所测型别符合率100%;B组肠道病毒所测型别仅有5个符合,符合率21%。结论基于VP4区基因序列的肠道病毒分型方法对A组肠道病毒能准确分型,对B组肠道病毒分型准确率欠佳,C组肠道病毒株较少,分型准确率有待进一步验证。
Objective To assess accuracy of the enterovirus typing method based on VP4 gene sequences and establish a rapid and simple method for typing enteroviruses. Methods VP4 genes of 126 enterovirus strains having been typed based on VP1 gene sequences were amplified by RT-PCR,and the amplification products were sequenced to obtain typing results by VP4 genes. Consistency between typing results obtained by VP4 genes and VP1 genes was assessed to determine accuracy of enterovirus typing by VP4 genes. Results Comparative analysis of VP4 gene sequences of 126 strains from 34 serotypes(8 serotypes of 37 enterovirus A,24 serotypes of85 enterovirus B,2 serotypes of 2 enterovirus C) showed a consistency rate of 100% between typing results by VP4 and VP1 gene regions for all serotypes of enterovirus A and C but a consistency rate of only 21% for only5 serotypes of enterovirus B. Conclusion Typing based on VP4 gene sequences is accurate for enterovirus A but insufficiently accurate for enterovirus B. Due to limited strains of enterovirus C in the study,the type accuracy awaits further verification.
