补肾温阳祛风散寒法对肾阳虚型围绝经期模型大鼠ER、VEGF、HSP70作用的研究

摘要 目的:探讨补肾温阳祛风散寒法对肾阳虚型围绝经期模型大鼠ER、VEGF、HSP70的影响及作用机制。方法:选用自然老化的围绝经期雌性SD大鼠,肌注氢化可的松构建肾阳虚型围绝经期大鼠模型,将模型大鼠随机分为更年汤组、教材方组、替勃龙组、生理盐水组;另选青年组、老年组SD雌性大鼠作为对照组。各组给予相应药物治疗15 d,于第16天处死大鼠,分离摘取双侧子宫,称重了解子宫湿重及子宫指数,采用病理切片观察大鼠子宫形态学变化,免疫组化及原位杂交检测方法观察大鼠子宫雌激素受体(ER)、血管内皮生长因子(VEGF)、热休克蛋白70(HSP70)及信使mRNA(ERmRNA、VEGFmRNA、HSP70mRNA)的阳性表达情况。结果:1更年汤组、教材方组、替勃龙组能明显增加子宫湿重、子宫指数,有统计学意义(P<0.05),更年汤组与替勃龙组、教材方组比较差异有统计学意义(P<0.05);2更年汤组、教材方组、替勃龙组能抑制子宫萎缩,有显著性差异(P<0.01),更年汤组与替勃龙组比较无统计学意义(P>0.05);3更年汤组、教材方组、替勃龙组能增加ER、ERmRNA、VEGF、VEGFmRNA阳性表达,有统计学意义(P<0.05),但教材方组的上调低于更年汤组及替勃龙组(P<0.05),替勃龙组与更年汤组之间比较无显著差异(P>0.05);4更年汤组、替勃龙组能增加HSP70及HSP70mRNA阳性表达,有统计学意义(P<0.05),更年汤组与替勃龙组比较无显著性差异(P>0.05)。结论:自拟更年汤可能通过上调ER、VEGF、HSP70蛋白及基因在子宫中的阳性表达而发挥雌激素样作用,从而达到改善病理状态下的子宫的形态结构及功能的目的。 Objective: To explore the influence of reinforcing kidney and warming Yang,expelling wind and cold method on ER,VEGF and HSP70 of perimenopause rats with kidney- Yang deficiency. And to explore its action mechanism. Methods: We chose natural aging perimenopausal female SD rats and injected hydrocortisone to build the rats model with deficiency of kidney-Yang. And then they were randomly divided into four groups: Gengnian Decoction group( A),textbook decoction group( B),Tibolone group( C) and normal saline group( D). A young group( E) and an elderly group( F) SD female rats were chosen as the control groups. After successful gavage of all rats of uncontrolled groups,we compared the positive expressions of uterine ER,ER mRNA,VEGF,VEGF mRNA,HSP70 and HSP70 mRNA in every group by Immunohistochemistry and in situ hybridization technique.Results: 1The Group A,B and C could up- regulate the uterine wet weight and uterus index. When compared with Group D and F and the difference was significant( P 0. 05). And the difference between Group A and Group B,C were significant( P 0.05). 2The Group A,B and C could check uterine atrophy,when compared with Group D and F,and the difference was significant( P 0. 01). And the difference between Group A and Group C had no significance( P 0. 05). 3The Group A,B and C could up- regulate the positive expression of uterine ER,ER mRNA,VEGF and VEGF mRNA( P 0. 05). The effect of Group A and C in promoting the expression were stronger than Group B while the difference between Group A and C had no statistical significance( P 0. 05). 4The positive expression of uterine HSP70 and HSP70 mRNA increased significantly in Group A and C and had statistical significance when compared with Group B,E and Group F. While the difference between Group A and C had no statistical significance( P 0. 05). Conclusion: The Gengnian Decoction could exert estrogen- like effects by up- regulating the expression of ER,ER mRNA,VEGF,VEGF mRNA,HSP70 and HSP70
