摘要
目的:建立青钱柳叶多糖柱前衍生化HPLC指纹图谱。方法:青钱柳多糖经三氟乙酸水解和1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化后,进行HPLC法分析。色谱分离采用Agilent ZORBAX XDB C18色谱柱(4.6mm×250mm,5μm),磷酸盐缓冲液(pH=6.8)-乙腈(83.5∶16.5)为流动相,流速1mL/min,检测波长250nm,柱温30℃,采用相似度分析、聚类分析和主成分分析对10批样品的图谱进行分析。结果:指纹图谱呈现8个共有峰,各批次样品相似度在0.9以上。结论:采用HPLC法建立多糖的指纹图谱操作简单快捷,可有效检测青钱柳多糖。
Objective:The HPLC finger print of pre-column derivatization of Cyclocarya paliurus polysaccharide was established.Methods:The Cyclocarya paliurus polysaccharide was hydrolyzed by trifluoroacetic acid and derived with 1-phenyl-3-methyl-5-pyrazolone(PMP).HPLC analysis was processed after that.Chromatographic separation using Agilent ZORBAX XDB C18 column(4.6 mm×250 mm,5μm),phosphate buffer(pH=6.8)-acetonitrile(83.5∶16.5)as mobile phase,flow rate 1 mL/min,detection wavelength was 250 nm,and the column temperature was 30°C.The maps of 10 batches were analyzed by similarity analysis,cluster analysis and principal component analysis.Results:The results showed that there were 8 common peaks in the fingerprint,and the similarity of each batch was above 0.9.Conclusion:HPLC establishing fingerprints of polysaccharides is simple and quick,and can effectively detect Cyclocarya paliurus polysaccharide.
