大补阴丸、牵正散及合方对帕金森小鼠脑神经元及其线粒体复合物酶活性的保护作用 被引量:11

Protective effects of Dabuyin Pills, Qianzheng Powder and both on neurons and activity of brain mitochondrial complex enzyme in mice with Parkinson's disease

摘要 目的:通过大补阴丸、牵正散及合方对帕金森病(PD)小鼠脑线粒体复合物酶Ⅳ(COX)活性及其亚基COXⅠmRNA相关作用的研究,探讨中药的神经保护机制。方法:采用腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)建立PD模型。75只C57BL/6雄性小鼠随机分为正常组、模型组、大补组、牵正组和合方组,每组15只。采用爬杆法测量小鼠行为学,应用HE染色观察中脑黑质病理学变化;免疫组织化学法检测TH阳性神经元数目;测定脑内COX的活性及其亚基COXⅠmRNA表达情况。结果:造模28d后,与模型组比较,3个中药治疗组能显著缩短PD小鼠的平均爬杆时间(P<0.05),增加中脑黑质神经元数目,改善神经元形态,并能明显减少TH阳性神经元丢失(P<0.05),提高COXⅠmRNA的表达(P<0.05),合方组可显著增加线粒体复合物酶Ⅳ活性(P<0.05)。结论:大补阴丸、牵正散及合方能提高PD小鼠脑COXⅠmRNA表达,且合方能改善PD小鼠脑线粒体复合物酶Ⅳ的活性,以维持正常线粒体功能,发挥神经保护作用。 Objective:To investigate the effects of Dabuyin Pills(DP),Qianzheng Powder(QP)and DP&QP on neurons,brain mitochondrial complexⅣ(COX)activity and its subunit cytochrome C oxidaseⅠ(COXⅠ)mRNA in MPTP-induced Parkinson’s disease mice.Methods:A model of Parkinson’s disease was established by intraperitoneal injection of MPTP.A total of75 C57 BL/6 male mice were randomly divided into the normal group,model group,DP group,QP group,and DP&QP group,there were 15 mice in each group.After 28 days,brains were taken after animals in each group were sacrificed.The ethological changes of rats were detected by climbing rod method.The pathological changes of substantia nigra neurons were observed by HE staining.The changes of dopaminergic neurons express in the substantial nigra and the brain activity of COX and the expression of its subunit(COX I)mRNA were measured.Results:After 28 days of model establishment,compared with the model group,the pole-climbing time shortened significantly in three therapy groups(P<0.05),increased the number of neurons in the midbrain,improve neuron morphology,TH positive neuron loss was significantly reduced(P<0.05),the expression of COX I mRNA increased(P<0.05),and the activity of brain mitochondrial complex enzyme IV were significantly increased in the DP&QP group(P<0.05).Conclusion:DP,QP and DP&QP can increase the expression of subunit COX I,and DP&QP can increase the activity of brain mitochondrial complex enzyme IV m RNA to maintain mitochondrial function and play a neuroprotective effect.
