紫草素抑制表皮生长因子促HaCaT细胞增殖的机制 被引量:10

Study on the mechanism of inhibition effect of shikonin on HaCaT cells proliferation induced by EGF

摘要 目的:研究紫草素抑制表皮生长因子(EGF)促HaCaT细胞增殖的机制及miR-21在其中的可能作用。方法:MTT法检测EGF、紫草素、miR-21模拟剂及miR-21抑制剂对HaCaT细胞增殖的影响;Western Blot法检测紫草素、EGF、miR-21模拟剂及miR-21抑制剂对HaCaT细胞PCNA表达的影响。结果:EGF浓度依赖性促进HaCaT细胞增殖;紫草素浓度依赖性抑制HaCaT细胞增殖;miR-21抑制剂、紫草素抑制EGF对HaCaT细胞的促增殖作用以及PCNA蛋白表达的上调作用,EGF、miR-21模拟剂部分逆转紫草素抑制HaCaT细胞增殖的作用以及对HaCaT细胞PCNA蛋白表达的下调作用。结论:紫草素可明显抑制EGF对HaCaT细胞的促增殖作用,其机制可能为通过抑制miR-21的表达,达到抑制HaCaT细胞增殖的作用。 Objective: To study the mechanism of shikonin in inhibiting HaCaT cells proliferation induced by EGF and the possible role of miR-21 playing in it. Methods: MTT assay was used to determine the proliferation of HaCaT cells incubated with EGF, shikonin, miR-21 mimic, miR-21 inhibitor. Western Blot was used to determine the protein expression of PCNA of HaCaT cells incubated with EGF, shikonin, miR-21 mimic, miR-21 inhibitor. Results: EGF promoted the proliferation of HaCaT cells in a dose-dependent manner, and shikonin inhibited the proliferation of HaCaT cells in a dose-dependent manner. miR-21 inhibitor and shikonin inhibited the effect of EGF on stimulating promoting proliferation of HaCaT cells and the protein expression level of PCNA. EGF and miR-21 mimic partially reverse the inhibiting effect of HaCaT cells promotion and PCNA expression by shikonin. Conclusion: Shikonincan inhibit the proliferation of HaCaT cells induced by EGF, and the mechanism may be the inhibition of the expression of miR-21, which suppresses HaCaT cell proliferation.
