摘要
目的:观察当归补血汤对糖尿病大鼠肾脏组织中氧化应激标志物(Nrf2、HO-1)表达的影响。方法:采用高脂饲料喂养联合低剂量链尿佐菌素腹腔注射的方法诱导糖尿病大鼠模型,将其分为模型组(DM)、格列齐特组(GL)、当归补血汤低剂量组(DL)、当归补血汤高剂量组(DH),另设正常对照组(N),每组10只,用药组分别给予格列齐特2.14mg·kg-1·d-1、当归补血汤1.78g·kg-1·d-1和7.14g·kg-1·d-1灌胃,模型组给予等量0.9%氯化钠溶液灌胃,连续处理8周后,检测各组大鼠肾脏组织中Nrf2、HO-1的含量,并观察各组大鼠肾脏组织的病理变化。结果:与N组相比,其余各组Nrf2、HO-1表达下降;病理切片显示DM组大鼠肾小球毛细血管基底膜不均匀增厚,系膜细胞增生,伴肾小管上皮细胞肥大,经格列齐特及当归补血汤处理后,上述指标及病理改变均减轻,DH组较GL组改善更明显(P<0.01),而DL组改善情况较GL组明显(P<0.01)。结论:当归补血汤能明显减轻糖尿病大鼠肾脏病理变化,与西药格列齐特相比,作用更显著,而且随着剂量的升高该作用增大,其机制可能与调控Nrf2、HO-1的表达,减轻氧化应激,增强抗氧化能力有关。
Objective: To investigate the effects of Danggui Buxue Decoction on the expression of oxidative parameters Nrf2 and HO-1 in kidney of Type 2 diabetes mellitus rats. Methods: Type 2 diabetes mellitus ratmodels were established by high fat diets and intraperitoneal injection with low-dose streptozotocin, and then randomly divided the rats into the diabetic model group(DM), the Gliclazide group(GL), the low-dose of Danggui Buxue Decoction group(DL) and the high-dose of Danggui Buxue Decoction group(DH). Except that, the normal group(N) was set. Rats in the DL and DH groups were administrated with1.78 g · kg-1 · d-1 and 7.14 g · kg-1 · d-1 of Danggui Buxue Decoction, respectively. Rats in the GL group was administrated with 2.14 mg · kg-1 · d-1 of Gliclazide. Rats in DM group were administrated with equivalent normal saline. After 8 weeks, rats were sacrificed and kidneys were harvested for pathological examination, gene and protein expression of Nrf2, HO-1 were detected by PCR and Western Blot, respectively. Results: Compared with N group, the level of Nrf2 and HO-1 were lower than that in other groups. Pathological changes were found in the kidney of rats from DM group, such as glomerular capillary basement membrane inhomogeneous thickening, mesangial cell proliferation, accompanied by renal tubular epithelial cells hypertrophy. Treatments with Gliclazide or Danggui Buxue Decoction could alleviated those pathological changes, the DH group were better than GL group in improving such changes(P<0.01), but the DL group was less effective than GL group in improving such changes(P<0.01). Conclusion: Danggui Buxue Decoction can effectively alleviate pathological changesin kidneys of type 2 diabetic mellitus rats and comparing with Gliclazide moreover, with the increase of the dose, the effect increases. The mechanism of which may relate with adjusting the expression of Nrf2 and HO-1.
