基于GC-MS对中医实热“上火”血清代谢特征的研究 被引量:12

Study on serum metabolic features of excessive heat by using gas chromatography-mass spectrometry

摘要 目的:全面地表征实热上火血清代谢谱的特异性变化,为实热上火机制的深入研究提供参考依据。方法:采用气相色谱-质谱联用对血清代谢物进行检测,结合OSC-PLS DA分析和t检验挖掘出实热上火发生显著变化的代谢物,关联相关代谢途径,解释实热上火状态下的人体响应变化。结果:实热上火与正常对照组血清代谢轮廓存在明显的差异,筛选并鉴定出其中18个差异代谢物。实热上火人群血清乳酸、油酸、亚油酸、甘油、酪氨酸和三羧酸循环中间物(柠檬酸、琥珀酸、苹果酸)等表达显著上调,而丝氨酸、苏氨酸、4-羟基脯氨酸等显著下调。结论:实热上火人群三羧酸循环、脂肪酸代谢、氨基酸代谢等发生特异性变化,鉴定的差异代谢物可为实热上火机制的研究提供依据。 Objective:To represent the specific changes of metabolic profile of excessive heat roundly,and provide a reference for the further study of the mechanism of excessive heat.Methods:The gas chromatography-mass spectrometry(GCMS) was used to test the serum metabolites of patients with excessive heat.The significant changes in metabolites and associated metabolic pathways of patients with excessive heat were excavated by using OSC-PLS DA analysis and t-test method,in order to explain the corresponding changes in the body under excessive heat.Results:There was a significant difference in the serum metabolic profile between the excessive heat and the normal control group,and 18 differential metabolites were screened and identified.The levels of serum lactic acid,oleic acid,linoleic acid,glycerol,tyrosine and tricarboxylic acid circulating intermediates(citric acid,succinic acid,malic acid) were significantly up-regulated,while serine,threonine,4-hydroxyproline and so on reduced significantly in patients with excessive heat.Conclusion:The differential metabolites in tricarboxylic acid cycle,fatty acid metabolism,amino acids can provide the basis for the study of the mechanism of excessive heat.
