摘要
目的:制备包裹磁性纳米粒子(Fe_3O_4)和抗肿瘤药物(吉西他滨,GEM)的磁性白蛋白纳米球,并研究其联合磁流体热疗对人肝癌SMMC 7721细胞生长的抑制作用。方法和结果:将包封在纳米球中的Fe_3O_4置于交变磁场中时,仍具有良好的磁响应性。Fe_3O_4白蛋白纳米球(平均粒径180 nm)热动力学试验显示,Fe_3O_4可以作为加热源使纳米球升温并达到稳定的温度(46℃)。采用MTT法检测GEM/Fe_3O_4白蛋白纳米球联合磁流体热疗和GEM化疗对肝癌细胞株SMMC 7721的抑制作用结果显示,GEM包裹在纳米球中可持续缓慢的释放,与载GEM白蛋白纳米球组和Fe_3O_4白蛋白纳米球联合交变磁场热疗组相比较,GEM/Fe_3O_4白蛋白纳米球联合交变磁场热疗组抗肿瘤作用增强。结论:GEM/Fe_3O_4白蛋白纳米球的热化疗协同增效有望应用于肝癌的治疗中。
Objective: To study the effect of nanosized magnetic albumin nanospheres encapsulating magnetic nanoparticles (Fe3O4 ) and antitumor drugs (Gemcitabine, GEM) in combination with magnetic fluid hyperthermia on human liver cancer SMMC 7721 cell growth. Methods: Nanosized magnetic albumin nanospheres encapsulating magnetic nanoparticles ( Fe3O4 ) and antitumor drugs ( Gemcitabine, GEM ) were designed. Results: AlthoughFe3O4 had been encapsulated in nanospheres, when exposed to an alternating magnetic field, the magnetic albumin nanospheres still had a good magnetic response with an average particle size of 180 nm. Thermodynamic tests showed that iron oxide could be used as a heating source and lead the nanospheres to reach a stable temperature (46 ℃ ). The release results in vitro indicated that nanospheres had an obvious effect of sustained release of GEM.The antitumor efficacy of the GEM/Fe3O4 albumin nanospheres combined with magnetic fluid hyperthermia on liver cancer cell line SMMC7721 was examined by MTT assay. Compared with nanospheres entrapping GEM group and nanospheres entrapping Fe3O4 combined with MFH group, the GEM/Fe3O4 albumin nanospheres exhibited enhanced antitumor efficacy. Conclusion: GEM/Fe3O4 albumin nanospheres have promising applications in liver cancer chemohyperthermia treatment.
