丹龙醒脑方对脑缺血再灌注大鼠海马区神经干细胞增殖及Frizzled1、Dvl1、CyclinD1表达的影响 被引量:9

摘要 目的:探讨丹龙醒脑方对局灶性脑缺血再灌注模型大鼠海马区神经干细胞增殖,以及Frizzled1、Dvl1、Cyclin D1表达的影响。方法:将80只雄性SD大鼠随机分为假手术组、模型组、依达拉奉组、丹龙醒脑方7.4g/kg组和丹龙醒脑方14.8g/kg组,采用线栓法制备局灶性脑缺血再灌注模型,再灌注1d、3d和7d采用改良m-NSS法进行神经功能缺损评分,7d后取大鼠缺血侧海马组织,采用免疫荧光法检测海马区Brdu阳性细胞率;采用RT-q PCR法、Western Blot法分别检测Frizzled1、Dvl1、Cyclin D1 mRNA和蛋白表达。结果:1再灌注1d,与假手术组比较,其余各组神经功能缺损评分明显升高,而各组间差异无统计学意义;再灌注3d、7d,与模型组比较,依达拉奉组、丹龙醒脑方7.4g/kg组、丹龙醒脑方14.8g/kg组评分明显降低。2与假手术组比较,各组Brdu阳性细胞率增大,Frizzled1、Dvl1、Cyclin D1mRNA及蛋白的表达明显增强;与模型组比较,依达拉奉组、丹龙醒脑方7.4g/kg组、丹龙醒脑方14.8g/kg组Brdu阳性细胞率增大,Frizzled1、Dvl1、Cyclin D1mRNA及蛋白的表达明显增强。结论:丹龙醒脑方能改善神经缺损症状,促进脑缺血后海马区NSCs增殖,其机制可能与通过上调缺血再灌注后海马区Frizzled1、Dvl1表达水平,从而促进Wnt/β-catenin信号通路下游靶基因Cyclin D1的表达水平有关。 Objective: To investigate the effects of Danlong xingnao recipe on proliferation of neural stem cells( NSCs) in hippocampus and the expressions of Frizzled1,Dvl1,Cyclin D1 with cerebral ischemia-reperfusion injury modle rats. Methods: 80 male SD rats were randomly divided into sham group,cerebral ischemia-reperfusion injury( model) group,edaravone group and Danlong Xingnao recipe( DLXNR) 7. 4g / kg group,DLXNR14. 8g / kg group. The focal cerebral ischemia reperfusion injury models were prepared by suture method,1d,3d,and 7d after reperfusion,conduct neural function defect scale in m-NSS,and 7d after reperfusion took the brain tissue ischemia side of rats with reperfusion. Brdu immunofluorescence method was used to record the hippocampus dentate gyrus Brd U positive cells number; The hippocampus Frizzled1,Dvl1,Cyclin D1 mRNA and protein expressions were determined respectively by RT-q PCR method and Western Blot method. Results:11d after reperfusion,Other groups were improved obviously than sham group in m-NSS,but there were no statistically significant difference between the each groups( P 0. 05); 3d or 7d after reperfusion,Compared with model group,edaravone group,DLXNR 7. 4g / kg group and DLXNR 14. 8g / kg group were lowed obviously( P 0. 05,P 0. 01); there were no statistically significant difference in other groups,( P 0.05). 2 Compared with sham group,the Others were improved obviously than sham group in Brdu positive cells rate and the expression of Frizzled1,Dvl1 and Cyclin D1 protein and mRNA( P 0. 05,P 0. 01); Compared with model group,edaravone group,DLXNR 7. 4g / kg group and DLXNR 14. 8g / kg group improved obviously in Brdu positive cells rate and the expression of Frizzled1,Dvl1 and Cyclin D1 protein and mRNA( P 0. 05,P 0. 01). Conclusion: Danlong Xingnao recipe can improve the neural functional recovery and promote the proliferation of NSCs in hippocampus after cerebral ischemia. One of the mechanism may be to improve the expressions of Frizzled1,Dvl1 in hip
