人鼻咽癌多药耐药细胞株CNE2/ADM的建立及生物学特性研究 被引量:1

摘要 目的建立人鼻咽癌多药耐药细胞株CNE2/ADM,并研究其生物学特性。方法以人鼻咽癌细胞株CNE2为亲本细胞,采用阿霉素(ADM)浓度梯度递增法诱导建立多药耐药细胞株CNE2/ADM。通过观察细胞形态学变化,药物敏感性试验,测定生长曲线、群体倍增时间、细胞周期及P-糖蛋白(P-gp)功能和表达情况,评价CNE2/ADM的生物学特性。结果与CNE2细胞相比,CNE2/ADM耐药细胞异形明显,并对ADM在内的4种化疗药物产生了耐药性,增殖速度减慢,群体倍增时间延长(P<0.05),G0/G1期细胞增多,S期、G2/M期细胞减少(P<0.05),P-gp的药物泵出功能增强、表达水平升高。结论成功建立了人鼻咽癌多药耐药细胞株CNE2/ADM,其具有典型的多药耐药表型,可作为进一步研究鼻咽癌多药耐药机制及筛选逆转剂较为理想的细胞模型。 Objective To establish a human nasopharyngeal carcinoma(NPC)multidrug-resistance cell line CNE2/ADM and to investigate its biological features.Methods Adriamycin resistance was induced in human NPC cell line CNE2 by continuous exposure to gradually increasing doses of adriamycin.Drug sensitivity,cell morphology,cell growth curves and the population doubling time,cell cycle distribution,the function and expression level of P-gp were investigated to determine the biological features of CNE2/ADM cell line.Results Compared with CNE2,the CNE2/ADM cell line was with significant heteromorphosis.It showed cross-resistance to four drugs such as adriamycin,the population doubling time was longer(P〈0.05)and the cell number of the G0/G1-phase was increased.Meanwhile,that of the S- phase and G2/M-phase decreased(P〈0.05),the function of P-gp was enhanced(P〈0.05)and the expression level of P-gp was up-regulated.Conclusion The MDR cell line CNE2/ADM is established successfully.CNE2/ADM cell line shows the typical multidrug resistance phenotype.It is a good model for exploring the mechanism of MDR and the selection of reversal agents.
