摘要
【目的】评价表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)对小鼠骨性关节炎的治疗作用。【方法】取C57BL/6小鼠36只,随机分为对照组、模型组和EGCG组,每组12只,通过韧带切除制造小鼠骨性关节炎模型。模型制作成功后连续给药,腹腔注射25 mg/kg EGCG,1次/d。3组小鼠分别于治疗4、8周后分批处死,采用骨性关节炎软骨病理变化评价系统评价关节软骨的病变,取小鼠膝关节软骨,切片后进行免疫组化染色,测量蛋白聚糖和Ⅱ型胶原染色平均灰度值用于评估关节软骨中蛋白多糖和Ⅱ型胶原的含量,计算基质金属蛋白酶-13(matrix metalloproteinase-13,MMP-13),聚蛋白聚糖酶(a disintegrin-like and metalloprotease with thrombospondin type I motifs-5,ADAMTS-5)阳性细胞百分率。运用实时定量PCR测定关节软骨中1-甲基环丙烯(1-methylcyclopropene,MCP-1)、趋化因子受体2(chemokine receptor2,CCR-2)、白细胞介素1β(interleuk in-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)RNA表达。【结果】骨性关节炎小鼠经EGCG治疗4、8周后,OARSI评分与模型组相比显著降低(P<0.01),关节软骨中蛋白聚糖、Ⅱ型胶原蛋白表达与模型组相比显著升高(P<0.01),MMP-13、ADAMTS-5阳性细胞百分率显著降低(P<0.01);治疗8周后,关节软骨中MCP-1、CCR-2、IL-1β、TNF-αm RNA表达与对照组相比显著减少(P<0.01)。【结论】EGCG能够缓解小鼠骨性关节炎的病理过程。
【Objective】To evaluate the effect of epigallocatechin- 3- gallate(EGCG) in the treatment of mouse osteoarthritis.【Methods】A total of 36 C57BL/6 mice were randomly divided into 3 groups: control group, model group and EGCG group, 12 mice per group. The mouse model of osteoarthritis was made via ligament exsection. The mice in EGCG group were injected intraperitoneally with EGCG(25 mg/kg) once a day after successful modeling. Mice in 3 groups were respectively sacrificed after 4w and 8w treatment. The lesions of articular cartilage were evaluated via OARSI. The levels of proteoglycans and type II collagen as well as the positive-cell percentage of MMP-13 and ADAMTS-5 in mouse articular cartilages were detected with immunohistochemistry. The m RNA expression levels of MCP-1, CCR-2, IL-1β and TNF-α in articular cartilages were determined with real-time quantitative PCR.【Results】After4w and 8 w treatment, the OARSI scores and positive-cell percentage of MMP-13 and ADAMTS-5 in EGCG group were significantly lower than those in model group(P0.01); the expression levels of proteoglycans and type II collagen in EGCG group were obviously higher than those in model group(P0.01). After 8 w treatment, the m RNA expression levels of MMP-13, CCR-2, IL-1β and TNF-αin EGCG group were significantly lower than those in model group(P0.01).【Conclusion】EGCG can alleviate the pathological changes of mouse osteoarthritis.
