摘要
目的:研究AGXT2L1是否能够影响肝细胞癌细胞系的运动、迁移和侵袭。方法:设计并构建AGXT2L1的靶向siRNA,验证敲低效果后,采用划痕愈合实验观察细胞的二维运动能力,采用Transwell实验观察细胞的三维迁移能力,采用带基质胶的Transwell实验观察细胞的侵袭能力。结果:与对照组相比,干扰AGXT2L1的表达可以显著抑制两种肝细胞癌细胞系(97H和HCCLM3)的运动、迁移和侵袭能力。结论:AGXT2L1具有调控肝细胞癌细胞运动、迁移和侵袭的功能,说明AGXT2L1可能在肝细胞癌的转移过程中发挥了重要作用;AGXT2L1是治疗肝细胞癌的潜在靶标。
Objective:To investigate the role of AGXT2L1 in motility,migration and invasion of hepatocellular carcinoma.Methods:siRNA targeting AGXT2L1 was designed,synthetized and transfected into the cells.After the validation of efficiency of knock-down,the motility of cells was measured by wound healing assay;the migration of the cells was measured by transwell assay;the invasion of the cells was measured by transwell with matrigel assay.Results:Compared with the control group,after the knock-down of AGXT2L1,the ability of motility,migration and invasion of 97 Hand HCCLM3cells both significantly reduced(P〈0.05).Conclusion:AGXT2L1can modulate motility,migration and invasion of hepatocellular carcinoma cells and it may be a potential target in hepatocellular carcinoma treatment.
