摘要
目的本研究旨在进一步研究阐明MicroRNA-421相对表达量与乳腺癌发生、发展及预后的关系。方法我院乳腺外科行乳腺癌手术切除患者的病例样本共计96例（从2012年1月～2015年1月）,对96例乳腺癌病例采用逆转录实时定量聚合酶链反应（RT-PCR）的方法对MicroRNA-421的相对表达量进行检测,运用Kaplan-Meier的生存曲线对肿瘤切除术后患者的生存情况进行分析。结果 Micro RNA-421相对表达量与患者的年龄、性别、分化、瘤组织大小等并不存在相关性（t=1.52,P=0.32;t=0.42,P=0.75;t=1.28,P=0.94;t=-1.22,P=0.12）;而远处转移方面,远处转移组的MicroRNA-421相对表达量明显下调（t=2.93,P=0.01〈0.05）,而无远处转移组的相对表达量与正常组比较,差异无统计学意义（t=1.36,P=0.98〉0.05）,MicroRNA-421相对表达量明显下调与远处转移的风险有关;生存分析方面,42例MicroRNA-421相对表达量明显下调的患者中生存时间为（37.34±10.90）个月,而MicroRNA-421高表达的患者生存时间为（54.23±12.43）个月,两组比较差异具有统计学意义（P〈0.05）,MicroRNA-421表达的高低与预后有着一定的相关性。结论 MicroRNA-421相对表达量水平与乳腺癌的远处转移及预后有着相关性,MicroRNA-421的下调可能有潜在抑癌基因的作用,可作为乳腺癌诊断和预后的新型标志物,需要进一步的研究验证。
Objective To further investigate the relationship between MicroRNA-421 relative expression levels and breast cancer metastasis and prognosis. Methods The total 96 samples were all came from our breast surgery depart ment, and all cases in this real-time quantitative reverse transcription polymerase chain reaction（RT-PCR） methods relative expression of MicroRNA-421＇s the amount of testing, using Kaplan-Meier survival curves for survival of patients after tumor resection were analyzed. Results There was no correlation of Micro RNA-421 relative expression and the patient＇s age, sex, degree of differentiation, tumor size, and so on（t=1.52, P=0.32; t=0.42, P=0.75; t=1.28, P=0.94;t=-1.22, P =0.12）;and in distant metastasis,the relative expression of distant metastasis of MicroRNA-421 was sig nificantly reduced（t =2.93, P=0.010.05）, the differences of without distant metastasis difference relative expression levels compared with the normal group were not statistically significant（t=1.36, P=0.980.05）, significantly reduced MicroRNA-421 was related to the risk of relative expression and distant metastasis; In survival analysis, 42 cases MicroRNA-421 relative expression was significantly lower in patients with survival time of（37.34 ±10.90） months, while patients with high expression of MicroRNA-421 survival time was（54.23±12.43） months, the differences were statistically significant（P〈0.05）, MicroRNA-421 expression level and the prognosis had some relevance. Conclusion MicroRNA-421 relative expression levels are related to breast cancer metastasis and prognosis, the relative expression of MicroRNA-421 as a tumor marker in breast cancer diagnostic and prognostic is needed to verify for further research.
