摘要 目的考察60Co辐照灭菌对黄芩饮片的主要色谱峰及含菌量的影响。方法选择5、10 k Gy 2种辐照剂量对黄芩饮片进行辐照;采用HPLC法建立黄芩特征图谱,比较辐照前后特征图谱中主要峰的峰面积变化率、红外光谱的变化情况;通过微生物限度检测法检测辐照前后黄芩饮片细菌和霉菌、大肠埃希菌及大肠菌群含菌量的变化情况。结果 2个剂量辐照后黄芩饮片微生物限度均达到卫生标准要求;红外光谱无明显变化;特征图谱中主要6个峰的含量均有明显变化,其中5 k Gy辐照后主要峰面积变化率均大于5%,10 k Gy辐照后主要峰面积变化率均大于30%。结论黄芩饮片不建议采用60Co射线辐照的方法灭菌。 Objective To investigate the effect of60 Co ray on the fingerpoints,infrared spectrum and microbialcontent of Scutellariae. Methods Scutellariae was irradiated with 5 and 10 k Gy. The fingerpoints of Scutellariae were detected by HPLC. The change rate of the peak area and infrared spectrum before and after irradiation were compared. The total amount of bacteria and mould,esherichia coli and coliforms living in the materals before and after irradiation were compared by microbial limit test. Results The microbial content of Scutellariae irradiated by either 5 or 10 k Gy reached the standard of the Chinese Pharmacopoeia. The infrared spectrum of Scutellariae did not significantly change,but the six main peak areas of Scutellariae changed obviously. After 5 k Gy irradiation,the change rate of the peak area was more than 5%. After 10 k Gy irradiation,the change rate was above 30%. Conclusion It is not advisable to use this method of sterilization for Scutellariae.
