苯肾上腺素对131碘致大鼠腮腺早期放射内损伤的作用及NAMPT表达的影响 被引量:2

摘要 目的研究苯肾上腺素(phenylephrine,PE)对131碘(^(131)I)致大鼠腮腺早期放射内损伤的作用及烟酰胺磷酸核糖转移酶(nicotinamide phosphoribosyltransferase,NAMPT)表达的影响。方法 Wistar大鼠随机分四组:空白对照组、单纯苯肾上腺素组(腹腔注射苯肾上腺素5 mg/kg)、单纯^(131)I组(腹腔注射^(131)I 0.5m Ci/100g)和苯肾上腺素预处理组(给予^(131)I前半小时腹腔注射苯肾上腺素),分别于给予^(131)I后1、7和14天取腮腺,显微镜下观察各组腮腺组织的形态学变化;免疫组织化学法检测NAMPT在腮腺的分布及表达。结果 ^(131)I导致腮腺腺泡发生空泡样变性、萎缩及核固缩,而苯肾上腺素预处理组上述细胞变性现象明显减轻。空白对照组NAMPT主要表达于浆液性腺泡细胞及导管细胞的胞浆和细胞核,单纯^(131)I组NAMPT的表达较空白对照组显著减少,而PE预处理组NAMPT的表达较单纯^(131)I组明显增加;单纯PE组NAMPT的表达较空白对照组无明显差异。结论苯肾上腺素可以减轻^(131)I所致的腮腺早期放射内损伤,其机制可能与苯肾上腺素诱导NAMPT表达上调有关。本研究将为探讨涎腺放射内损伤的分子新机制及防治放射性涎腺损伤提供新思路。 Objective To investigate the effect of phenylephrine on the alteration of morphology and the expression of nicotinamide phosphoribosyltransferase(NAMPT) after ^131I radiation in rat parotid gland. Methods 108 Wistar rats were randomly divided into four groups: the control group(n=9); phenylephrine alone group(n=9,phenylephrine was injected intraperitoneally,5mg/kg);^131I alone group(n=9,^131I was injected intraperitoneally,0.5m Ci/100g); and ^131 I with phenylephrine pretreatment group(phenylephrine was injected intraperitoneally 30 min prior to administration ^131I). After administration of ^131I 1,7,and 14 days,hematoxylineosin staining observed the morphology change of parotid gland under microscope; Immunohistochemical assay was used to detect the distribution and expression of NAMPT in parotid gland. Results ^131I resulted in vacuolization,atrophy and pyknotic nuclei in acinar cells,whereas pretreatment phenylephrine markedly alleviated acinar cell atrophy in parotid gland. NAMPT was mainly expressed in cytoplasm and nucleus of serous acinar and duct cells,^131I significantly decreased the expression of NAMPT in serous acinar and duct cells,however,phenylephrine increased the expression of NAMPT nearly to the control level. Conclusion These results suggest that phenylephrine could reduce the damage induced by ^131I in parotid gland,which underlying mechanism might be related to the NAMPT upregulation by phenylephrine.
