摘要 目的观察系统性红斑狼疮(SLE)患者外周血T淋巴细胞p53基因表达情况及探讨其与DNA甲基化的相关性。方法收集34例SLE患者与23例健康对照者,均取外周血T淋巴细胞,分别用5-甲基胞嘧啶抗体与流式细胞仪检测DNA甲基化状态和反转录-聚合酶链反应(real time RT-PCR)分析p53mRNA表达情况。结果 SLE患者DNA甲基化水平活动期(8.50±1.42)与正常人(11.31±1.34)相比,差异有统计学意义(P<0.01)。SLE患者p53基因表达活动期(1.15±0.33)与正常人(0.81±0.27)相比,差异也有统计学意义(P<0.01)。SLE患者p53基因的表达与狼疮疾病活动指数(SLEDAI)正相关(P<0.01);而与DNA甲基化水平没有明显的相关性(P>0.05)。结论 p53基因在外周血T淋巴细胞中的异常表达与SLE的发病相关,p53基因的异常表达在SLE表观遗传学发病机制的作用有待进一步研究。 Objective To analyze association between the expression of p53 - mRNA and DNA methylation status in T cells from patients with systemic lupus erythematosus (SLE). Methods The DNA methylation status in T cells from 34 SLE patients and 23 healthy controls was assessed by the specific monoclonal antibodies to 5 - methylcytosine(5 -mc) and analyzed quantitatively by flow cytometry. Real time RT- PCR was applied to analyze p53 -mRNA levels in T cells from the patients and controls. Results Active SLE patients had significant DNA hypomethylation than controls ( P 〈 0.01 ). Active SLE patients had significantly higher levels of p53 - mRNA than controls ( P 〈 0.01 ) and the level of p53 - mRNA was positive correlated with the SLE - DAI but with no correlation between the levels of p53 mRNA and DNA methylation. Conclusion Abnormal expression of p53 - mRNA plays an important role in the pathogenesis of SLE. The mechanism of higher expression of p53 - mRNA in patients with SLE should be studied further.
