摘要
本试验旨在研究含黄素单氧化酶3(Flavin-Containing Monooxygenase 3,FMO3)基因T329S突变在河北太行鸡群体中的分布。本研究根据Gen Bank中公布的鸡FMO3基因序列设计引物,采用PCR-RFLP方法对517只河北太行鸡FMO3基因频率及基因型频率进行检测。结果显示,该位点在河北太行鸡群体中处于Hardy-Weinberg平衡状态(P>0.05);等位基因A、T频率分别为0.9023、0.0976;AA、AT、TT基因型频率分别为0.8201、0.1644、0.0155。结果表明,河北太行鸡群体中鱼腥味综合症易感基因型频率不高,可以通过PCR-RFLP的方法予以剔除。
The objective of the study was to detect the distribution of Flavin-Containing Monooxygenase 3(FM03) T329S mutation in Taihang Chickens of Hebei. In the experiment, primers were de- signed according to FM03 gene sequences in GenBank database. PCR-RFLP technique was used for detecting the T329S polymorphism site of FM03 gene in 517 Taihang Chickens. The result of c^2 test showed that the polymorphic site of FM03 gene was fit with Hardy-Weinberg equilibrium in Taihang Chickens of Hebei (P〉0. 05), The allele frequencies of A, T were 0.9023, 0.0976; the frequencies of AA, AT and TT genotype were 0.8201, 0.1644 and 0.0155, respectively. The research result has showed that the frequency of fishy taint genotype was generally low and the susceptible individuals could be easily detected by PCR-RFLP method in the Taibang Chicken of Hebei.
