摘要
以拟南芥中控制硫苷生物合成相关基因家族GS-Elong的全长CDS及蛋白序列为参考序列,在甘蓝(Brassica oleracea L.)基因组数据库中进行BLASTN和BLASTP检索,得到具备Score≥100、E-value≤10-10和Coverage≥70%条件的甘蓝注释基因作为拟南芥GS-Elong位点基因的候选同源基因,并进一步对其进行同源性和基因结构比较及聚类分析。结果显示,在甘蓝GS-Elong位点发现4个旁系同源基因(Bol020647、Bol017070、Bol037823和Bol017071)。甘蓝和拟南芥GS-Elong位点同源基因在核酸水平上的一致性为66.1%~84.3%;甘蓝4个旁系同源基因间的一致性在62.0%~83.7%之间。甘蓝和拟南芥GSElong位点基因在外显子数目、大小方面高度一致,但Bol017071和Bol037823分别较拟南芥GS-Elong位点基因少3个和1个外显子。系统进化及共线性分析初步确定,甘蓝中存在2个BoMAM1(Bol017070和Bol020647)和2个Bo MAM3(Bol017071和Bol037823)。
Using the full CDS and protein sequences of Arabidopsis glucosinolate biosynthesis related GS-Elong gene family as references,the candidate homologous genes with Score ≥100,E-value ≤10-10 and Coverage ≥ 70% were identified from genome database of Brassica oleracea L. through BLASTN and BLASTP. Then,the homology and structures of these genes were compared and the clustering analysis was conducted. The results indicated that there were four candidate GS-Elong genes( Bol020647,Bol017070,Bol037823 and Bol017071) in Brassica oleracea. The homologous GS-Elong genes in Brassica oleracea and Arabidopsis shared 66. 1% ~ 84. 3% nucleotide sequence identity,and four paralogs GS-Elong genes in Brassica oleracea shared 62. 0% ~ 83. 7% nucleotide sequence identity. The Brassica oleracea and Arabidopsis GS-Elong genes were highly consistent on the number and size of exon,but the two Brassica oleracea GS-Elong genes,Bol017071 and Bol037823,had three and one exon less respectively than those of Arabidopsis.The phylogenetic and collinearity analyses showed that there were two Bo MAM1( Bol017070 and Bol020647)and two Bo MAM3( Bol017071 and Bol037823) in Brassica oleracea.
