摘要
目的探讨白藜芦醇拮抗游离脂肪酸诱导的体外培养的胰岛B细胞凋亡的可能机制。方法将10μmol/L白藜芦醇和500μmol/L棕榈酸同时作用于体外培养的胰岛B细胞。共分为3组:空白组(不含棕榈酸)、棕榈酸组(PA组,含500μmol/L棕榈酸的脂性培养基)、白藜芦醇组(Res组,含10μmol/L白藜芦醇+500μmol/L棕榈酸)。以流式细胞术检测细胞凋亡,RT-PCR检测凋亡相关蛋白Bax和Bcl-2 mRNA水平。结果与PA组相比,Res能显著降低B细胞凋亡率(95.82%±1.94%vs.11.03%±1.08%,P<0.01),降低Bax mRNA(0.185±0.005 vs.0.161±0.003,P<0.01),升高Bcl-2 mRNA(0.004±0.001 vs.0.008±0.001,P<0.01)。结论白藜芦醇能拮抗棕榈酸诱导的胰岛B细胞凋亡,其机制可能与调节凋亡相关蛋白有关。
Objective To investigate the mechanism of resveratrol in inhibiting apoptosis of B cells induced by palmitic acid( PA),a free fatty acid,in vitro. Methods B cells were treated with 10 μmol / L resveratrol +500 μmol / L palmitic acid in vitro,and were divided into three groups: blank group( excluding free fatty acids),PA group( 500 μmol / L palmitic acid lipid medium),resveratrol group( 10 μmol / L resveratrol + 500 μmol / L palmitic acid),respectively. The percentage of apoptotic cells was calculated by flowcytometry. Bax and Bcl-2 mRNA were measured by RT-PCR. Results At 24 h,compared with PA group,resveratrol could significantly decrease B cells apoptosis percentage( 95. 82% ± 1. 94% vs. 11. 03% ± 1. 08%,P〈0. 01) and Bax mRNA( 0. 185 ± 0. 005 vs. 0. 161 ±0. 003,P〈0. 01),elevate Bcl-2 mRNA( 0. 004 ± 0. 001 vs. 0. 008 ± 0. 001,P〈0. 01). Conclusion Resveratrol can significantly inhibit B cell apoptosis induced by palmitic acid in vitro. The mechanism may be related to the regulation of resveratrol on some apoptosis protein.
