从胚胎上皮细胞基因表达谱变化分析肺与大肠相表里内涵 被引量:28

摘要 目的:比较胚胎发育3个时期肺与结肠上皮细胞基因表达水平的相似性,为肺、肠同源发生提供依据,丰富"肺与大肠相表里内涵"。方法:应用c DNA微阵列技术对胚胎肺组织与结肠组织基因表达情况进行检测和比较,分离2种组织上皮细胞,抽提细胞总RNA、扩增、Cy3染色后,与牛全基因组芯片杂交,荧光信号扫描,以Agilent feature extraction软件分析,筛选同源表达基因,进一步对同源表达基因进行GO分析和pathway分析;以qRT-PCR验证芯片结果。结果:在胚胎肺、结肠不同时期,两者上皮细胞基因表达均有不同程度的相似性;GO分析表明在两种胚胎上皮细胞发育过程中,3个时期上皮细胞生物过程的表达基因符合率分别为15%、27%、53%,其中比例较高的有生物调节、细胞分化、细胞周期等;3个时期上皮细胞分子功能表达基因符合率分别为19%、27%、24%,其中比例较高的包括转移酶活性、激酶活性和酶调节活性;3个时期上皮细胞组分表达基因符合率分别为66%、46%,23%,比例较高的为非膜结合细胞器、胞外区、大分子复合物组分等;pathway分析显示m TOR信号通路、PI3K/Akt、wnt通路有明显相似性,共涉及14个相似基因表达。结论:胚胎发育3个时期,肺与肠基因表达均存在不同程度的相似性,早期二者相似基因表达主要体现在上皮细胞形态发育上;中晚期则主要在于二者功能上。 Objective: To compare the gene expression levels of the lung cells and the colon epithelial cells to explain connotation of how lung and intestine come out homogonously. Method: The embryonic lung tissue and colon tissue gene expression were examined with cD NA microarray technology and were compared by separating two kinds of epithelium cells,extracting,amplifying and Cy3 dying the total RNA expressions. After the bovine genome microarray hybridization and fluorescent signal scan,with Agilent feature extraction software analysis,the homologous genes wer found out. By GO analysis and pathway analysis of the homologous genes,the microarray results was verified. Results: In the embryonic lung,colon at different times,both epithelial cell gene expressions share certain degrees of similarity. GO analysis showed that in both embryonic epithelial cell development,the similarity rates of the expression of the gene in epithelial cells of the biological processes at three stages were 15%,27%,53% respectively,among which the similarity in biological regulation,cell differentiation,cell cycle were more significant. The similarity rates of molecular function of the epithelial cells at three stages were 19%,27%,and 24% respectively,among which the similarity of transferase activity,kinase activity and enzyme regulator activity were more significant. The similarity rates of component genes of the epithelialcells at three stages were 66%,46%,23% respectively,among which the similarity of non-membrane-bound organelles,extracellular region macromolecular complex components were more significant. Pathway analysis suggested that there exists obvious similarity among mT OR signaling pathway,PI3 K / Akt and wnt pathway,with a total of 14 similar gene expressions being quite similar.Conclusion: The embryonic development of lung and intestinal gene expression at three stages share great similarity,which is mainly reflected in the epithelial cell morphology development at early stage and is reflected in cell functions at late st
