摘要
目的:探讨大鼠急性脊髓损伤(ASCI)后,应用免疫组化染色观察神经元凋亡抑制蛋白(NAIP)、存活素(Survivin)在ASCI后的表达和脊髓损伤的关系,并进一步探讨脊髓损伤后神经细胞凋亡抑制相关蛋白表达的重要意义。方法:取SD大鼠50只制作急性脊髓夹压伤动物模型,损伤后1、3、7天于损伤节段脊髓组织取材,NAIP、Survivin免疫组化染色,正常大鼠为正常对照组。计算机图像分析仪定量分析,观察急性脊髓损伤与NAIP、Survivin表达的相关性。结果:正常脊髓内NAIP、Survivin有微弱的表达,损伤后1、3、7天脊髓灰质、白质及室管膜细胞内均有阳性表达。NAIP的表达1D时最显著(P<0.05),表达强弱的顺序是:1天>3天>7天(P=0.202)。Survivin的表达3D时最显著(P<0.05),表达强弱的顺序是:3天>7天(P=0.011)>1天(P=0.015)。结论:哺乳类动物急性脊髓损伤后启动了以神经元凋亡抑制蛋白、存活素表达为特征的自我保护与修复机制,且神经元凋亡抑制蛋白、存活素表达的程度与脊髓损伤的程度呈正相关。
Objective: To observe the relationship between the expressions of neuronal apoptosis inhibitory protein(NAIP) and survivin in rats and spinal cord injury after acute spinal cord injury(ASCI) by immunohistochemical dyeing, further explore important meaning of NAIP expressions. Methods: Fifty SD rats were chosen and prepared as the models with ASCI, the tissues were taken out from spinal marrow of the injured segments in one day,three and seven days after ASCI, NAIP and Survivin were observed after immunohistochemical dyeing, normal rats as normal control group. The relativity between ASCI and the expressions of NAIP and Survivin were observed in quantitative analysis with computer imagining analyser. Results: NAIP and Survivin were found slight expressions in normal spinal marrow; they had positive expressions in gray matter, white matter and ependymal cells of spinal cord in one day, three and seven days after ASCI. NAIP expressed the most significantly in 1D(P〈0.05), the expressions in the order of 1D〉3Ds〉7Ds(P=0.202). Survivin expressed the most significantly in 3Ds(P〈0.05), its expressions in the order of 3Ds〉7Ds(P=0.011)〉1D(P=0.015). Conclusion: The mechanism of self-defense and restoration featured by the expressions of NAIP and Survivin is started in the body of mammals after ASCI, the expressions of NAIP and Survivin are positively related to the degrees of spinal cord injury.
