鞘内注射不同剂量神经妥乐平对大鼠脊髓神经的影响 被引量:2

Effects of intrathecal different doses of neurotropin on spinal neurotoxicity in rats

摘要 目的观察鞘内注射不同剂量神经妥乐平对大鼠神经行为学、脊髓组织的病理变化及c-fos蛋白表达的影响,探讨大鼠鞘内注射神经妥乐平的脊髓神经毒性。方法清洁级雄性SD大鼠,体重220—250g,取鞘内置管成功的大鼠60只,随机分为4组(n=15):鞘内注射神经妥乐平1.2NU/ml10μl(Ⅰ组);鞘内注射神经妥乐平0.6NU/ml10μl(Ⅱ组);鞘内注射神经妥乐平0.3NU/ml10μ1(Ⅲ组);鞘内注射生理盐水10μl(Ⅳ组)。于鞘内置管前1d及鞘内给药后1、3、7d分别行平面翻正试验并测定左后足底机械刺激缩足阈值(PWT),于给药后1、3、7d每组随机选取5只大鼠,取腰膨大处脊髓组织,苏木素一伊红(HE)染色进行组织病理学观察,并采用免疫组织化学法测定给药后1d脊髓背角c—fos蛋白表达水平。结果各组大鼠(每组5例)各时间点平面翻正试验均为阳性。Ⅰ、Ⅱ、Ⅲ和Ⅳ组大鼠鞘内置管前1d、鞘内给药后1、3、7d左后足底PwT分别为I(14.81±0.24)、(14,77±0.23)、(14.84±0.25)、(14.79±0.30)gl;[(14.58±0.29)、(14.80±0.24)、(14.87±0.26)、(14.58±0.30)g];f(14.54±0.28)、(14.61±0.31)、(14.54±0.28)、(14.81-t-O.24)g]和[(14.90±0.27)、(14.58±0.30)、(14.87±0.26)、(14.81±0.31)g],与鞘内置管前1d比较,给药后1、3、7d,各组大鼠左后足底PWT差异无统计学意义(P〉0.05)。与Ⅳ组比较,Ⅰ、Ⅱ、Ⅲ组大鼠给药后各时间点PWT差异无统计学意义(P〉0.05)。光镜下观察,各组大鼠给药后1、3和7d,脊髓组织均无明显病理变化。Ⅰ、Ⅱ、Ⅲ、Ⅳ组大鼠鞘内给药后1d脊髓背角e—fos蛋白表达的阳性细胞数分别为(14.95±1.02)、(15.75±1.19)、(15.64±1.27)、(15.19±1.25)个,与Ⅳ组比较,Ⅰ、Ⅱ、Ⅲ组给药后 Objective To investigate the spinal neurotoxicity of intrathecal different doses of neu- rotropin in rats. Methods Sixty male SD rats weighing 220 - 250 g were randomly divided into 4 groups (n=15 each): neurotropin - treated groups [1.2 NU/ml, (group I ), 0.6 NU/ml (group II), 0. 3 NU/ml (group III) 10 μl respectively] and intrathecal injection of saline solution 10 μl (group IV ). The surface righting and the paw withdrawl mechanical threshold (PWT) on the 1st day before cathether- ization of the lumbar subarachnoid space, and 1 st, 3rd and 7th day after the intrathecal injection were ob- served. Five rats were randomly selected in each group at 1 st, 3rd and 7th day after the intrathecal injec- tion, the lumbar enlargement of spinal cord was taken out, the hematoxylin and eosin (HE) staining was used to observe histopathological changes, and the immunohistochemical method was applied to detect the expression levels of c - los protein at first day after the treatment. Results PWT at 1st day before the in- trathecal injection, and at 1st, 3rd and 7th day after the intrathecal injection in groups I,II,III and IV was (14.81 ±0.24), (14.77 ±0.23), (14.84 ±0.25) and (14.79 ±0.30) g, (14.58 ±0.29), (14.80 ± 0.24), (14.87±0.26)and (14.58±0.30) g, (14.54±0.28), (14.61±.31), (14.54 ±.28) and (14.81 ±0.24) g, and (14.90 ±-0.27), (14.58 ±.30), (14.87 ±0.26) and (14.81 ±.31) g, respeetively. As compared with that on the 1 st day before the intratheeal injection, there were no significant differences in the PWT at 1st, 3rd and 7th day after the intratheeal injection among all groups. As corn-pared with group IV, there were no significant differences in the PWT at 1st, 3rd and 7th day after the in- trathecal injection in groups I , II and III (P 〉 0. 05).
