摘要
目的:建立测定四取代四苯基卟啉衍生物(TLTAPP)含量的方法。方法:采用反相高效液相色谱法。色谱柱为Waters Sun Fire^(TM)C_(18),流动相A为乙腈-水(含0.1%三氟乙酸,95∶5,V/V)、流动相B为乙腈-水(含0.1%三氟乙酸,5∶95,V/V)(梯度洗脱),流速为1.0 ml/min,柱温为30℃,检测波长为418 nm,进样量为20μl。结果:TLTAPP检测质量浓度线性范围为1.55~95μg/ml(r=0.999 4);检测限和定量限分别为1.1 ng和3.8 ng;精密度、稳定性、重复性试验的RSD<1%;加样回收率为99.00%~101.60%,RSD=0.89%(n=9)。结论:该方法简单、快速、准确、专属性强,可用于TLTAPP的含量测定。
OBJECTIVE:To establish a method for the content determination of tetra-substituted tetraphenylporphyrin derivative(TLTAPP). METHODS:RP-HPLC was performed on the column of Waters Sun Fire^TMC18 with the mobile phase A of acetonitrile-water(containing 0.1% trifluoroacetic acid,95 ∶ 5,V/V)and mobile phase B of acetonitrile-water(containing 0.1% trifluoroacetic acid,5 ∶ 95,V/V)(gradient elution)at a flow rate of 1.0 ml/min;the column temperature was 30 ℃,the detection wavelength was 418 nm and the injection volume was 20 μl. RESULTS:The linear range of TLTAPP was 1.55-95 μg/ml(r=0.999 4);the detection limit and quantitation limit were 1.1 ng and 3.8 ng;RSDs of precision,reproducibility and stability tests were lower than1%;recovery was 99.00%-101.60%(RSD=0.89%,n=9). CONCLUSIONS:The method is simple,rapid,accurate and specific,and can be used for the content determination of TLTAPP.
