咳嗽变异性哮喘肝郁肺虚病证结合动物模型的建立和评价 被引量:8

摘要 目的:探索CVA肝郁肺虚病证结合大鼠动物模型的构建方法。方法:模拟中医病因,以幼龄大鼠肺虚、阴液亏虚,烟熏致肺虚,长期激怒致肝郁犯肺建立肝郁肺虚中医证候模型;模拟西医病因病理,卵蛋白氢氧化铝致敏,辣椒素激发,建立西医大鼠CVA模型。观察大鼠一般状态、自发活动情况、咳嗽反应、肺支气管病理组织形态、支气管肺泡灌洗液(BALF)嗜酸性粒细胞计数。结果:肝郁肺虚模型组大鼠体重增长缓慢(P<0.01),自发活动增多(P<0.01),咳嗽次数增加(P<0.01);支气管管腔狭窄,黏膜增厚,肺泡腔增大,黏液分泌增多,大量嗜酸性粒细胞浸润(P<0.01);与模型组比较,肝肺同治组各项观测指标均有明显改善(P<0.01)。结论:模拟传统中医病因、西医病因病理建立CVA动物模型;以整体观念、微观验证综合评价中医CVA动物模型;以方测证,肝肺同治反证CVA动物模型。多因素复合作用可成功复制CVA大鼠病证结合动物模型。 Objective: To establish the animal model of liver depression and lung deficiency syndrome of cough variant asthma. Methods: By simulating the transitional Chinese medicine( TCM) etiology,the young rats were smoked to lung deficiency as their Qi and Yin deficiency in lung in natural. And then irritate them for a long time to lead to the liver invading the lung syndrome,and thus the animal model of liver depression and lung deficiency was established. By simulating the western medicine etiology pathology,the young rats were sensitized by egg protein aluminum hydroxide and inspired by capsaicin,and then establish the animal model of cough variant asthma rat. Observe the rat general state,spontaneous activity,cough response,bronchial lung pathology organization form and bronchoalveolar lavage fluid( BALF) eosinophil count. Results: The rat weight increased slowly in liver depression and lung deficiency group( P〈0. 01),spontaneous activity increased( P〈0. 01) and cough frequency increased( P〈0. 01). The bronchial lumen stenosis,mucosal thickening,alveolar space increased,mucus secretion increased,a large number of eosinophil infiltration( P〈0. 01).Compared to the model group,the observation indexes of treatment on lung and liver group were obviously improved( P〈0. 01). Conclusion: To establish the animal model of cough variant asthma rat by simulating TCM etiology and western medicine etiology. To evaluate the animal model of cough variant asthma rat by the holism and microcosmic. To prove the model of cough variant asthma rat by taking a square to test certificate and treatment on lung and liver in opposite. Multifactor composite effects can be used successfully in replication of animal model of cough variant asthma rat with combination of disease and syndrome.
