摘要
目的研究槐耳清膏对人胃癌MKN-45细胞在裸鼠体内生长和转移的抑制作用,并探讨其作用机制。方法采用BALB/c裸鼠建立人胃癌MKN-45细胞淋巴结转移模型,随机分为槐耳清膏组、空白对照组,每组20只。建模后,每5天测量裸鼠荷瘤并计算肿瘤相对体积(RTV);用药5周后处死裸鼠,Real-time q PCR检测肿瘤组织Wnt、β-catenin、E-cadherin、N-cadherin和VEGF表达水平。结果槐耳清膏组末次测得的足垫瘤RTV为(662.75±450.66)mm3,明显小于空白对照组的(1611.85±611.83)mm3(P<0.01)。槐耳清膏组淋巴结转移率8/20(40%)与对照组15/20(70%)相比,差异有统计学意义(P<0.05)。槐耳清膏组肿瘤组织Wnt、β-catenin、N-cadherin和VEGF的△CT值分别为(11.36±1.32)、(11.31±1.76)、(12.82±2.22)、(13.22±1.52),均高于对照组的(9.48±2.07)、(9.37±1.88)、(7.74±2.64)、(11.04±1.61),而E-cadherin的△CT值(8.26±2.39)低于对照组的(10.64±1.48)(P<0.05)。结论槐耳清膏可抑制人胃癌MKN-45细胞在裸鼠上的生长及转移,其机制可能与抗血管生成和阻碍上皮间质转化(EMT)过程有关。
Objective To study the effects of trametes robiniophila on the growth and metastasis of human gastric cancer cell MKN-45 in nude mice,and to explore the mechanism.Methods BALB/c nude mice were inoculated with MKN-45 cells to establish the lymph node metastasis model of human gastric carcinoma.After the model was successfully established,40 mice were randomly divided into control group and trametes robiniophila group(n=20 in each group).After the mass formed,the caliper was used to measure the tumor size every 5 days.The relative tumor volume(RTV)was calculated.After 5 weeks,the nude mice were killed for determination of Wnt,beta-catenin,N-cadherin,VEGF m RNA expression by Real-time q PCR.Results After 5 weeks of drugs intervention,the RTV in trametes robiniophila group were less than that in control group(662.75 ±450.66mm3 vs1611.85±611.83mm^3,P〈0.01).The lymph node metastasis rate of trametes robiniophila group was 8/20(40%),lower than that in control group 15/20(75%)(P〈0.05).Compared with control group,the △CT value of Wnt,β-catenin,N-cadherin,and VEGF in trametes robiniophila groupwere higher(11.36±1.32 vs 9.48±2.07,11.31±1.76 vs 9.37±1.88,12.82±2.22 vs 7.74±2.64,13.22±1.52 vs 11.04±1.61),the △CT value of E-cadherin were lower(8.26±2.39 vs10.64±1.48),with all P〈0.05.Conclusion Trametes robiniophila can inhibit the growth and lymphatic metastasis of human gastric cancer in nude mice,which might be related with anti-angiogenesis and setting the epithelial-mesenchymal transition(EMT)process back.
